Whole Blood Immunoassay Based on Centrifugal Bead Sedimentation

Abstract: BACKGROUND:Centrifugal "lab on a disk" microfluidics is a promising avenue for developing portable, lowcost, automated immunoassays. However, the necessity of incorporating multiple wash steps results in complicated designs that increase the time and sample/reagent volumes needed to run assays and raises the probability of errors. We present proof of principle for a disk-based microfluidic immunoassay technique that processes blood samples without conventional wash steps.

“…This process also separated the beads from the red blood cells in the case that they are present, and provided a washing step for the beads. 109 Koh et al performed a similar sedimentation step with only one bead type for the detection of botulinum toxin and achieved a lower limit of detection of 0.09 pg/mL. 110 …”

“…Towards this goal, we have developed multiplexed immunoassays that can be implemented in a portable microfluidic platform developed by our group previously. 29,30 The sensing method used was sandwich immunoassays performed using silica beads (Figure 1). High specificity antibodies (capture antibodies) were covalently attached to the silica beads using a PEG linker.…”

“…An innovative modification in centrifugal microfluidics has been the implementation of bead-based immunoassays using density-media centrifugation (15). This allows bodily fluids, such as WB and saliva, to be analyzed directly because cells are separated by centrifugation from the analytes, which are captured on antibody-bearing beads.…”
