We skip to work: alternative splicing in normal and malignant myelopoiesis

Abstract: Alternative splicing expands the transcriptome thereby promoting protein diversity. It governs critical cellular processes such as differentiation, proliferation and apoptosis in a tissue-specific manner. Aberrant splicing consequent to mutations in splicing factors and disruption of isoform ratios in key regulatory genes provides an important contribution to the pathogenesis of the myelodysplastic syndromes and myeloid leukemia. We review here the central role of alternative splicing in regulating myelopoiesi… Show more

“…Interestingly, even if aberrant alternative splicing plays a relevant role in AML [57] and m 6 A modification has an established role in regulating alternative splicing, mRNA translation has been identified as the main regulatory step deregulated by m 6 A in AML [51,52,53]. Therein, the YTHDF1 reader should mediate most of the observed phenotype in AML.…”

“…Changes in expression levels and specific binding of the hnRNP A/B protein affect this developmental switch 15 . Since then, alternative splicing has emerged as a more widespread phenomenon 1618 . The muscleblind-like protein 1 (MBNL1) is a sequence-specific splicing factor that undergoes extensive alternative splicing during differentiation 17 .…”
