This study evaluated the effect of adhesive systems and application strategies on dentin hydraulic conductance (HC). The buccal enamel was removed from bovine incisors to simulate laminate cavity preparations. After removing the roots and the coronal pulp, the buccal dentin was treated with EDTA solution (0.5 M) for 5 minutes, rinsed, ultrasonicated for 12 minutes and connected to a permeability device. HC of the specimens was measured at 10 psi (n = 5). Permeability was measured before and after bonding procedures using G-Bond (GB), Clearfil Tri-S Bond (CTS), Hybrid Coat (HY), Bond Force (BF), Adper Easy Bond (AEB) Silorane (SI), Clearfil SE Bond (CSE) and Adper Scotchbond Multi-Purpose (SMP) adhesives systems, which were applied following three strategies: 1) according to the manufacturers' instructions; 2) two coats of all-in-one self-etching adhesives (GB, CTS, HY, BF, AEB) or priming step plus two coats of bonding resin for the other systems (SI, CSE and SMP); and 3) a thin layer of a flowable composite applied over one coat of all-in-one self-etching adhesives or primed surface for SI, CSE and SMP adhesives. No significant difference was observed among the application modes concerning their ability to reduce HC. None of the adhesives showed complete sealing (100%) of the bovine tooth dentin. SI exhibited lower HC than SMP, however, they were not significantly different from the other systems. The results suggest that all systems tested result in an HC reduction of more than 90%. The wet bonding technique seemed to be more sensitive for dentin sealing.

Testing endodontic sealers on human umbilical vein endothelial cells

AbstractThe toxic potential of the endodontic sealers ingredients, especially the unreacted monomer, that can irritate the periapical tissue and interfere with the healing process, thus having a negative impact on the biocompatibility of the material. The aim of this study was to evaluate the influence of three experimental endodontic sealers on cells viability in vitro. Human umbilical vein endothelial cells (HUVEC) were used. The experiments were done with solid samples and extracts of sealers in artificial saliva and water. The experiments evaluated the cytotoxicity of the residual monomers that resulted from the tested composites. The decrease in cell viability was quantified by colorimetric measurement of formazan. The components of the sealers dissolved in artificial saliva and water were determined by high performance liquid chromatography (HPLC). The HUVEC are a novelty for testing the endodontic sealers biocompatibility, with certain advantages compared to other cell types used in the literature, e.g. HELA cells, fibroblasts. The data showed that cytotoxicity was directly linked with the unreacted monomer — 2-hydroxyethyl methacrylate (HEMA) present in these composites. Two of the three formulations had little or no cytotoxic effect, which makes them suitable for further testing in order to be used in endodontic treatment.

Ten-year observation of dentin bonding durability of 4-META/MMA-TBB resin cement —a SEM and TEM study

This study aimed to assess dentin bond durability of 4-META/MMA-TBB resin cement over ten years, by evaluating the tensile bond strength, and SEM and TEM observations. Tensile bond strength of Super Bond C&B (SB) to bovine dentin was evaluated at 1 day and after 10 years. The mode of failure after debonding was observed by SEM. Interfacial ultrastructure and nanoleakage was observed by TEM at the baseline and after 10 y. The tensile bond strength significantly dropped after 10 y. The failure pattern shifted from cohesive failure in resin towards adhesive failure or cohesive failure in dentin. TEM observation revealed degradation of both resin and collagen networks within the hybrid layer and nanoleakage at the base of the hybrid layer after 10 y. The bond strength of SB to dentin significantly decreased, and the hybrid layer degraded, while the overlying hydrophobic resin layer showed little disintegration over 10 y.
