Wasp venom immunotherapy changes IgG antibody specificity

Abstract: The immunodominance pattern of IgG epitopes recognized on wasp venom antigens by sera from wasp allergic patients changes soon after initiating rush VIT. Further studies will indicate whether, instead of measuring IgG titres, this marked change could be used as the basis of a new test for monitoring the outcome of VIT.

“…We have not been able to find significant alterations in IgG or IgG4 avidity by aeroallergen SIT [14, 29], which have been shown in insect venom studies [30, 31]. This supports that IgG4 contributes to the blocking activity by simple quantity rather than altered quality [32, 33].…”

“…A change in IgE specificity has also been documented during grass pollen immunotherapy [ 22]. Furthermore, these early results emphasize that, even when established, antigenic immunodominance of IgG antibody is not a permanent state and can be reversed by intervention on the immune system, for example, through the injection large amounts of antigens [ 23–25].…”

“…Similarly, Michils et al [7] reported that an increase in specific IgG during venom SIT was associated with a rapid change in antibody specificity. Interestingly, the allergen binding of pooled IgG fractions from allergic individuals was inhibited to a greater extent by sera from other allergic patients than by sera from SIT patients, despite higher antibody titres in the latter.…”

“…However, a correlation between increases in IgG4 titres and clinical improvement has not always been found [4,5]. One explanation for this is provided by increasing evidence that suggests that the biological activity as well as the quantity of IgG4 antibodies is altered during successful SIT [6][7][8]. SIT-induced IgG4 antibodies thus have IgE blocking activities that are not necessarily reflected in the antibody titre but require biological assays for detection [6, [9][10][11][12].…”
