Volume Reconstruction Techniques Improve the Correlation Between Histological and in vivo Tumor Volume Measurements in Mouse Models of Human Gliomas

Abstract: Assessment of therapy efficacy using animal models of tumorigenic cancer requires the ability to accurately measure changes in tumor volume over the duration of disease course. In order to be meaningful, in vivo tumor volume measurements by non-invasive techniques must correlate with tumor volume measurements from endpoint histological analysis. Tumor volume is frequently assessed by endpoint histological analyses approximating the tumor volume with geometric primitives such as spheroids and ellipsoids. In thi… Show more

“…The intratumoral distribution of CMDiI-labeled NSCs was assessed by fluorescence microscopy. The tumor volumes were estimated using the formula for ellipsoids and expressed as means ± SE, as described previously (27). Immunohistochemistry was done with the Vectastain Elite ABC kit (Vector Laboratories).…”

“…C6 cells (10 mL of a 10 6 =mL cell suspension) in an ultra-low gelling temperature agarose were injected into the cortex to a 3 mm depth at a rate of 2 mL=min. 5,6 A waiting time of 2 min was implemented following injection and bone wax was put in the burr-hole to prevent any reflux. The wound was sutured and covered with surgical glue.…”

“…There is a high demand for noninvasive imaging techniques to determine precisely the onset of tumor growth and to characterize growth over time, especially in spontaneous or induced murine tumor models such as WAP-T mice. Therefore, techniques have been applied that enable imaging and exact monitoring of tumor growth and progression in small animals, such as magnetic resonance imaging (MRI 7 ), ultrasound, [8][9][10] computed tomography (CT 11 ) and nuclear imaging. 12,13 Each modality possesses a unique combination of advantages and disadvantages that affect their selection for use in a particular study.…”
