Synthetic fungicides are a common treatment for post-harvest diseases in fruits, although demand is increasing for alternatives such as biopesticides. The aim of this work was to investigate the presence of naturally occurring antifungal compounds in Brazilian specimens of the weed Conyza canadensis. Two compounds [(4Z)-lachnophyllum lactone and (4Z,8Z)-matricaria lactone] were isolated and their antifungal activities were evaluated against eight postharvest disease fungi. Aspergillus niger, Cladosporium sp., and Penicillium digitatum proved susceptible to the treatment (minimum inhibitory concentrations varying from 32 to 64 µg mL -1 ). These fungi are common postharvest pathogens of fruits and vegetables, causing rots and secreting mycotoxins. A green and sustainable extraction method employing pressurized hot water was developed and optimized (100 °C, 4 × 1 min cycles). Yields of 1.46 and 0.24 mg g -1 were obtained for (4Z)-lachnophyllum lactone and (4Z,8Z)-matricaria lactone, respectively. The extract could be applied directly to fruits and vegetables in postharvest treatments.

Introduction:The present work aims to study the chemical composition of Pteridium aquilinum (L.) Kuhn essential oil and its antibacterial activity against three important phytopathogenic Gram-negative bacteria: Erwinia amylovora, Pectobacterium carotovorum subsp. carotovorum, and Pseudomonas savastanoi pv. savastanoi. Methods: The chemical composition of P. aquilinum L. essential oil produced by hydrodistillation was determined by gas chromatography-mass spectrometry. The antibacterial activity was tested using disk diffusion method and by determination of minimum inhibitory concentration values. The major components were linalool (10.29%), carvacrol (8.15%), benzaldehyde (5.95%), 2-undecanone (5.32%), and cuminaldehyde (4.57%). Results: The essential oil tested revealed a powerful antibacterial effect against all tested strains, with inhibition zone diameters ranging from 32.0 ± 0.58 to 33.7 ± 0.88 mm. Discussion: P. aquilinum EO contained 32.86% of oxygenated monoterpenes, which are known for their very powerful antimicrobial activities. The minimum inhibitory concentration values showed that P. aquilinum essential oil has very strong activity against E. amylovora (0.625 μl/ml), followed by P. carotovorum subsp. carotovorum (2.50 μl/ml) and P. savastanoi pv. savastanoi (5.00 μl/ml). The results obtained could contribute to the development of new potential agents for the control of bacterial diseases.

Sesquiterpenes are characteristic components and important quality criterions for agarwood. Although sesquiterpenes are well-known to be biosynthesized by sesquiterpene synthases (TPSs), to date, only a few TPS genes involved in agarwood formation have been reported. Here, two new TPS genes, namely, TPS9 and TPS12, were isolated from Aquilaria sinensis (Lour.) Gilg, and their functions were examined in Escherichia coli BL21(DE3), with farnesyl pyrophosphate (FPP) and geranyl pyrophosphate (GPP) as the substrate of the corresponding enzyme activities. They were both identified as a multiproduct enzymes. After incubation with FPP, TPS9 liberated β-farnesene and cis-sesquisabinene hydrate as main products, with cedrol and another unidentified sesquiterpene as minor products. TPS12 catalyzes the formation of β-farnesene, nerolidol, γ-eudesmol, and hinesol. After incubation with GPP, TPS9 generated citronellol and geraniol as main products, with seven minor products. TPS12 converted GPP into four monoterpenes, with citral as the main product, and three minor products. Both TPS9 and TPS12 showed much higher expression in the two major tissues emitting floral volatiles: flowers and agarwood. Further, RT-PCR analysis showed TPS9 and TPS12 are typical genes mainly expressed during later stages of stress response, which is better known than that of chromone derivatives. This study will advance our understanding of agarwood formation and provide a solid theoretical foundation for clarifying its mechanism in A. sinensis.
