Viral danger signals control CD1d de novo synthesis and NKT cell activation

Abstract: The nonpolymorphic CD1 molecules present lipid antigens to T cells. In myeloid DC humans express five different CD1 proteins (CD1a-e; the corresponding CD1 genes are designated CD1A-E). A role for CD1d-restricted NKT cells in the control of virus infections has been delineated from clinical observations, mouse models and viral evasion mechanisms targeting CD1d. How NKT cells are activated by virus infections is unclear. We found that human myeloid DC differentially regulate CD1 antigen presentation in response… Show more

“…27 Another subset of DCs, plasmacytoid DCs (pDCs), secretes type1 interferon and supports mDCs in promoting iNKT cell activation. 28 Furthermore, activated iNKT cells also enhance acquired immunity. Thus, human iNKT cells bridge the innate and adaptive immune responses by interacting with both DCs and NK cells, and they play crucial roles in controlling viral infection and tumor cells.…”

“…pDCs cooperate with mDCs to promote antiviral activity by activating iNKT cells. 28,33 Therefore, we subsequently analyzed the surface expression level of DC-activation markers, such as CD40, CD80, CD86, and HLA-ABC on the mDCs and pDCs of 9 HDs, 7 ACs, 9 HAM/TSP patients, and 3 ATL patients (supplemental Figure 3). CD40 and CD80 expression on both circulating mDCs and pDCs (data not shown) and CD86 expression on pDCs (supplemental Figure 3A) were negligible in all subjects.…”

“…However, how is the expansion of NKT early after SIV infection generated? Viral danger signals through the type I IFN pathway trigger NKTactivation by myeloid dendritic cells (48,49). DNA and RNA viruses induce type I IFN through TLR7 and TLR9, and, in particular, HIV has been shown to induce type I IFN (50).…”

“…Blockade of NKG2D on nonclassical NKT cells prevented hepatitis B virus-induced acute hepatitis in a CD1d-dependent manner (63). Upon infection of human DC, low titers of herpes simplex virus (HSV) upregulated cell surface CD1b and CD1d expression, whereas high titers did the opposite, suggesting that CD1-mediated antigen presentation by DC is a target for HSV immune evasion (50,51). This idea is supported by effects shown on CD1d recycling by HSV (65) or Kaposi's sarcoma-associated herpesvirus (56), and HSV-2 virus titers in NKT-cell-deficient mice are elevated in a genital herpes mouse model (4).…”
