Reactive RNA probes are useful for studying and identifying RNA-binding proteins. To that end, we designed and synthesized chloroacetamide-linked 7deaza-ATP which was a good substrate for T7 RNA polymerase in in vitro transcription assay to synthesize reactive RNA probes bearing one or several reactive modifications. Modified RNA probes reacted with thiolcontaining molecules as well as with cysteine-or histidine-containing peptides to form stable covalent products. They also reacted selectively with RNA-binding proteins to form cross-linked conjugates in high conversions thanks to proximity effect. Our modified nucleotide and RNA probes are promising tools for applications in RNA (bio)conjugations or RNA proteomics.

A series of 7-substituted 7-deazaadenine and 5substituted cytosine 2'-deoxyribonucleoside triphosphates (dNTPs) were tested for their competitive incorporations (in the presence of dATP and dCTP) into DNA by several DNA polymerases by using analysis based on cleavage by restriction endonucleases. 7-Aryl-7-deazaadenine dNTPs were more efficient substrates than dATP because of their higher affinity for the active site of the enzyme, as proved by kinetic measurements and calculations.

Ein neu synthetisierter DNA-Baustein auf Basis von 2'-Desoxyuridin trägt in der 5-Position ein über eine Aminopropinylgruppe verknüpftes, push-pull-substituiertes Diaryltetrazol. Das damit modifizierte Oligonucleotid ermçglicht die postsynthetische Markierung mit einem Maleinimid tragenden Sulfo-Cy3-Farbstoff, N-Methylmaleinimid und Methylmethacrylat als Dipolarophile jeweils durch Bestrahlung mit Licht bei l exc = 365 nm (LED). Die experimentell bestimmte Geschwindigkeitskonstante beträgt (23 AE 7) m À1 s À1 und ist bemerkenswert groß gegenüber denen anderer kupferfreier, bioorthogonaler Reaktionen und ähnlich groß wie jene der kupferkatalysierten Cycloaddition von Aziden mit Acetylenen.Die postsynthetische Modifizierung von Nucleinsäuren ist eine inzwischen gut etablierte Methodik, die hauptsächlich als kupferkatalysierte Cycloaddition von Acetylenen mit Aziden durchgeführt wird. [1,2] Auch wenn die Markierung in lebenden Zellen durch kupferkatalysierte "Klick"-Reaktionen demonstriert wurde, ist es problematisch, sich dabei auf Kupfer zu verlassen, [3] denn selbst Spuren von Kupferionen als Verunreinigungen aus der postsynthetischen oder In-vivo-Modifizierung stellen wegen ihrer Cytoxizität eine deutliche Beeinträchtigung dar. [4] Deswegen konzentrieren wir [5] und andere uns darauf, [6][7][8][9][10][11] bioorthogonale und kupferfreie Alternativen zu entwickeln, um Nucleinsäuren zu modifizieren. Das schließt ringspannungsgetriebene [5] und andere 1,3-dipolare Cycloadditionen, [6] Diels-Alder-Reaktionen mit normalem [7] und inversem Elektronenbedarf, [8] reduktive Aminierungen, [9] Thiol-Alken-Additionen [10] und Suzuki-Miyaura-Kupplungen [11] mit ein. Es gibt zwei metallfreie, bioorthogonale Reaktionen, die eine gute Alternative bieten, weil ihre Geschwindigkeitskonstanten vergleichbar mit denen der kupferkatalysierten Azid-Acetylen-Cycloaddition sind (k % 10-200 m À1 s À1 ). [12,13] Dies sind erstens die Diels-Alder-Reaktion mit inversem Elektronenbedarf von Tetrazinen mit gespannten Alkenen (k % 1-10 4 m À1 s À1 ) [8,12,14] und zweitens die lichtinduzierte Cycloaddition von Tetrazolen mit aktivierten Alkenen (Photoklick-Reaktion, k % 60 m À1 s À1 ).
