Vasotocin and isotocin regulate aquaporin 1 function in the sea bream

Abstract: Aquaporins (AQPs) are specific transmembrane water channels with an important function in water homeostasis. In terrestrial vertebrates, AQP2 function is regulated by vasopressin (AVP) to accomplish key functions in osmoregulation. The endocrine control of aquaporin function in teleosts remains little studied. Therefore, in this study we investigated the regulatory role of vasotocin (AVTR) and isotocin (ITR) receptors in Aqp1 paralog gene function in the teleost gilthead sea bream (Sparus aurata). The complete… Show more

“…Methodology for the ex vivo opercular epithelia preparation followed our current methods. 45 Fish were anaesthetized with 2-phenoxyethanol (1 : 2000 v/v), sacrificed by decapitation and the cranium was cut longitudinally. The gills and other tissue remains were removed carefully and the epithelial skin covering the opercular bone were dissected out and transferred to fresh-gassed saline (99.7 : 0.3 O 2 /CO 2 ) with the following composition (all values in mM): NaCl, 160; MgSO 4 , 0.93; NaH 2 PO 4 , 3.0; CaCl 2 , 1.5; NaHCO 3 , 17.85; KCl, 3.0; glucose, 5.5; HEPES (pH 7.8), 5.0.…”

“…Vasopressin is important for water balancing via the control of antidiuresis and regulation of blood pressure and plays a role in influencing social behaviors such as territorial defense, aggression, and also pair bonding (58). Across vertebrates, oxytocin/vasopressin-like peptide homologs appear to have conserved physiological (9, 10) and behavioral functions (1115).…”

“…The seawater teleost rectum also plays a role in water absorption. For instance, in the sea bream (Sparus aurata) and Japanese eel (Anguilla japonica), fluid absorption by the rectum also occurs via aquaporins, but the rate of absorption is less than in the intestine (8,15,28,36). The rectum of the Gulf toadfish (Opsanus beta) also expresses intestinal ion transporters and enzymes involved in solute-coupled water absorption, such as SLC26a6, the basolateral electrogenic Na ϩ /HCO 3 Ϫ -cotransporter [NBCe1 (SLC4a4)], apical VHA, and cytosolic carbonic anhydrase (CA C ) (20,22,42,49).…”
