Vasoactive intestinal polypeptide and nitric oxide synthase distribution in the enteric plexuses of the human colon: an histochemical study and quantitative analysis

Abstract: (a) the right colon contains a lower number of neurons and of VIP- and NOS-positive ones than the left colon, and (b) VIP- and NOS-positive fibers are differently distributed in the inner and outer portions of the circular muscle.

“…In this study, a double-labeling immunofluorescent technique using antibodies was employed to determine the population of neuronal bodies in the enteric plexuses of the colon from chagasic patients with megacolon. Calretinin has been used to identify intrinsic primary afferent neurons (IPANs), cChAT and SP to identify excitatory motor neurons, NPY to identify interneurons, and NOS and VIP to identify inhibitory motor neurons in the human colon [10,[12][13][14].…”

“…NOS-IR myenteric plexus (MP) neurons were counted (twice by two different workers) in sections obtained from paraffin-embedded specimens which were first immunostained and further stained with haematoxylin-eosin (HE) to determine the MP neuron number per millimetre of section length (see Matini et al 1995) in rats sacrificed during the first week of postnatal life (P0, P3, P5, P7; two animals for each age group) and in adulthood (P30, P60, P90; two animals for each group of age). In order to ascertain whether the increase in NOS-immunoreactive neuron number occurred gradually or at a specific age, six 2-week-old rats and six 3-week-old rats were also studied.…”

“…A constitutive nitric oxide synthase is present in neurons (NOS-1). In the gut it is localized in the neurons of enteric ganglia and in their processes (1)(2)(3). The function of NO produced by NOS-1 is relaxation of the smooth muscle.…”
