Vascular graft healing: I. FTIR analysis of an implant model for studying the healing of a vascular graft

Abstract: FTIR analysis of sequential biopsy samples of the primary and replacement segments obtained from 70 to 80-cm long carotid-femoral bypass grafts implanted bilaterally in the dog indicate that the healing pattern is similar over the entire length of these grafts. Preliminary analysis of the spectra also indicates that the major developments in the formation of the biological layer appear to occur during the first 4 weeks after implantation. Collagen IV could be detected by both FTIR and antibody staining in the … Show more

“…13,14 Of interest is the increase in the 1034 cm Ϫ1 absorption relative to the 1076 cm Ϫ1 absorption, which would suggest the beginning of collagen IV formation in this mixture of saccharide-containing material. 5,10,11 Because collagen IV is an indicator of the presence of endothelial cells, 25 one would expect the detection (by histology techniques) of some endothelial cells on the luminal surface of the grafts. However, this was not the case.…”

“…There are also increases in the bands associated with collagen IV (1031 cm Ϫ1 band relative to the 1077 cm Ϫ1 band), 5,10,11 suggesting that some of these cellular membranes are from endothelial cells, because collagen IV has been reported to be a marker for endothelial cells. 25 Histologies did show that endothelial cells are present at this time point.…”

“…Samples of ePTFE segments, retrieved at 1 and 3 h; 3, 5, 7, 10, and 14 days; and 4 and 8 weeks postimplantation, were analyzed by FTIR‐ATR. Empirical rules coupled with a number of experimental and theoretical studies7, 9–24 were used to assist in IR band assignments for these spectra.…”

“…A small band at 2120 cm −1 , due to a combination of water vibrations,28, 29 can be used for the subtraction of water, because biochemical materials usually do not absorb at this wavelength. In addition, the development of ATR techniques for examination of surfaces has allowed the nondestructive analysis of human tissue 7, 9–12, 16–21. These studies have shown that one can determine the presence of various collagens (and other proteins), cholesterol, and cholesterol esters in human tissue.…”
