Effect of Tulbaghia violacea on the blood pressure and heart rate in male spontaneously hypertensive Wistar rats I. A. RAJITulbaghia violacea Harv. (Alliaceae) is a small bulbous herb which belongs to the family, Alliaceae, most commonly associated with onions and garlic. In South Africa (SA), this herb has been traditionally used in the treatment of various ailments, including fever, colds, asthma, paralysis, hypertension (HTN) and stomach problems. The aim of this study was to evaluate the effect of methanol leaf extracts (MLE) of T. violacea on the blood pressure (BP) and heart rate (HR) in anaesthetized male spontaneously hypertensive rats; and to find out the mechanism(s) by which it acts.The MLE of T. violacea (5 -150 mg/kg), angiotensin I (ang I, 3.1 -100 µg/kg), captopril (10 mg/kg), angiotensin II (ang II, 3.1 -50 µg/kg), losartan (30 mg/kg), phenylephrine T. violacea (60 mg/kg) and captopril (10 mg/kg) were injected intraperitoneally into iii some SHR for 21 days to investigate the chronic effect of these agents on plasma levels of aldosterone. The mean change, the mean of the individual percentage changes and the percentage difference (in mean) observed with each intervention was calculated and statistically analyzed using the Student's t test for significant difference (p < 0.05). The Microsoft Excel software was used for statistical analysis.T. violacea significantly (p < 0.05) reduced the systolic, diastolic, and mean arterial BP;and HR dose-dependently. In a dose-dependent manner, ang I, ang II, phenylephrine significantly (p < 0.05) increased the BP, while propranolol, muscarine and atropine reduced the BP. The increases in BP due to dobutamine were not dose-dependent. In a dose dependent manner, phenylephrine and propranolol reduced the HR, while dobutamine increased the HR. The effect of ang I, ang II, muscarine and atropine on HR were not dose-dependent; with both increases as well as decreases observed with ang I, and II and atropine, while decreases were seen with muscarine. Captopril produced significant (p < 0.05) reduction in BP which were not associated with any change in HR.The co-infusion of ang I with the MLE produced significant (p < 0.05) reduction in BP, which were not associated with significant changes in HR. ABSTRACT:Background: Visceral fat accumulation due to the decrease in estrogen levels, and gaining weight after menopause, increases the risk of metabolic syndrome and cardiovascular diseases in postmenopausal women; while physical activity at maximum fat oxidation intensity (Fatmax) and green tea consumption are recommended for increasing fat oxidation. The aim of this study was to evaluate the effect of short-term aerobic exercise and green tea consumption on maximum fat oxidation (MFO), Fatmax, body composition and lipid profile in sedentary postmenopausal women. Methods: 24 sedentary postmenopausal women were divided into two groups: exercise+placebo and exercise+supplement. Exercise tests were performed by gas analyzer device and blood samples were collected after 12 hours of fasting overnight in pre and posttest phases. The subjects had a daily intake of 1,200 mg of green tea or cellulose in capsule form. The training protocol contained two weeks of aerobic exercise at an intensity of about maximum fat oxidation which was performed four times a week and each session took 40 to 50 minutes. Results: After two weeks, the amounts of weight, body mass index, waist to hip ratio, average total carbohydrate oxidation and HDL were significantly decreased in both groups (p≤0.05). The peak oxygen consumption, maximum fat oxidation, Fatmax and the average total fat oxidation increased but were not significant (p>0.05). Triglyceride (P=0.003) and visceral fat levels (P=0.044) only declined significantly in exercise+supplement group and LDL levels (P=0.043) only increased in exercise+supplement, significantly. Fat percentage also decreased in two groups which was not significant (p>0.05). Abstract. The aim of this study was to evaluate the effect of hypertension and its reverse on serum nitric oxide (NO) concentration and endothelial permeability in two-kidney one-clip (2K1C) hypertensive rats. 28 male Wistar rats were divided into four groups: 1) 2K1C for 12 weeks; 2) sham-clipped for 12 weeks; 3) 2K1C for 12 weeks and unclipped for 12 weeks; 4) sham-clipped for 12 weeks and unclipped for 12 weeks. Blood samples were taken before experiment, 12 th week and 24 th week (in groups 3 and 4). Coronary vascular and aortic endothelial permeability were determined by extravasation of Evans blue dye method. Serum NO level was significantly lower in hypertensive group compare with sham group (4.21 ± 1.28 vs. 9.47 ± 1.34 µmol/l, respectively). Reversal of hypertension did not improve serum NO concentration in 2K1C group (4.21 ± 1.28 vs. 4.32 ± 1.34 µmol/l). Coronary vascular and aortic endothelial permeability were not different between hypertensive and normotensive groups and reversal of hypertension did not alter endothelial permeability. Lower serum NO concentration in 2K1C hypertensive rats even after reversal of hypertension suggested that in addition to NO, other mechanisms could be involved in surgical reversal of hypertension. Hypertension and its reverse did not change endothelial permeability at least in this model of hypertension.
