Variation in the IL1B, TNF and IL6 genes and individual susceptibility to prosthetic joint infection

Abstract: BackgroundProsthetic joint infection (PJI) is an important failure mechanism of total joint arthroplasty (TJA). Here we examine whether the particular genetic variants can lead to increased susceptibility to PJI development.ResultsWe conducted a genetic-association study to determine whether PJI could be associated with functional cytokine gene polymorphisms (CGP) influencing on innate immunity response. A case–control design was utilized and previously published criteria for PJI were included to distinguish b… Show more

“…A validated tool for screening patients for increased risk of PJI does not currently exist. Despite attempts to identify and stratify patients at risk of PJI [ 12 , 42 , 64 , 65 , 66 ] specific clinical algorithms are not routinely used. In addition, we have no data relevant for determining the potential costs associated with wide range usage of such a screening strategy.…”

“…Many inflammation-related genes are found to be associated with risk of CHD [ 9 , 10 ]. Interleukin 1, beta gene ( IL1B ) encodes the cytokine which exerts a wide range of inflammatory activities [ 11 ]. Genetic association between IL1B and CHD has been previously found [ 12 ].…”

“…In contrast, the effect of smoking on tissue blood flow and oxygenation at the time of surgery is biologically plausible. A recently demonstrated association between a polymorphism in the gene encoding interleukin-1␤ and a higher risk of PJI suggests an interesting new area of research in the era of individualized medicine (49).…”
