Background The lipopeptide herbicolin A (HA) secreted by the biocontrol agent Pantoea agglomerans ZJU23 is a promising antifungal drug to combat fungal pathogens by targeting lipid rafts, both in agricultural and clinical settings. Improvement of HA production would be of great significance in promoting its commercialization. This study aims to enhance the HA production in ZJU23 by combining fermentation optimization and strain engineering. Results Based on the results in the single-factor experiments, corn steep liquor, temperature and initial pH were identified as the significant affecting factors by the Plackett–Burman design. The fermentation medium and conditions were further optimized using the Box-Behnken response surface method, and the HA production of the wild type strain ZJU23 was improved from ~ 87 mg/mL in King’s B medium to ~ 211 mg/mL in HA induction (HAI) medium. A transposon library was constructed in ZJU23 to screen for mutants with higher HA production, and two transcriptional repressors for HA biosynthesis, LrhA and PurR, were identified. Disruption of the LrhA gene led to increased mRNA expression of HA biosynthetic genes, and subsequently improved about twofold HA production. Finally, the HA production reached ~ 471 mg/mL in the ΔLrhA mutant under optimized fermentation conditions, which is about 5.4 times higher than before (~ 87 mg/mL). The bacterial suspension of the ΔLrhA mutant fermented in HAI medium significantly enhanced its biocontrol efficacy against gray mold disease and Fusarium crown rot of wheat, showing equivalent control efficacies as the chemical fungicides used in this study. Furthermore, HA was effective against fungicide resistant Botrytis cinerea. Increased HA production substantially improved the control efficacy against gray mold disease caused by a pyrimethanil resistant strain. Conclusions This study reveals that the transcriptional repressor LrhA negatively regulates HA biosynthesis and the defined HAI medium is suitable for HA production. These findings provide an extended basis for large-scale production of HA and promote biofungicide development based on ZJU23 and HA in the future. Graphical Abstract

Aiming at the potential application of lipopeptide biosurfactant (BioS) in bioremediation, we studied its production by a novel, isolated strain of Bacillus sp. MI27. Using the experimental design methodology, a sucrose-based medium composed of 2% sucrose, 0.27% Na2HPO4, 0.2% ammonium sulfate, 0.02% NaCl, 0.02% CaCl2, 0.02% MgSO4, 0.001% MnSO4, 0.06% KH2PO4, 0.005% FeSO4 and 0.005% ZnSO4 was optimized. With this composition, strain MI27 produces 1.4 g/L with maximum surface tension (ST) reduction of 23 mN/m and a dispersion diameter of around 10 cm. Emulsifying and foaming activities have been also confirmed. The critical micelle concentration (CMC) value of about 120 mg/L with a maximal decrease of ST of 23 mN/m with a maximum dispersion and an emulsification index (EI) of about 12 cm and 45% at 1,000 mg/L. Moreover, the foaming capacity is about 80% at 1,000 mg/L stable over 1 h of incubation. Additionally, we studied the effect of different values of pH, temperature and salinities on MI27 BioS activity and stability. Obtained results showed interesting surface activities at extreme physicochemical conditions, especially at acidic and alkaline pH values, high and low temperatures and higher salinities. All these characteristics enable the possible application of BioS in water treatment biotechnology under diverse environmental conditions.

The application of halophilic biosurfactant-producing bacteria to remove the pollutants from textile effluent was investigated in the current study. Staphylococcus sciuri subsp. rodentium strain SE I isolated from Sambhar Salt Lake, Rajasthan showed the highest emulsification activity of 65 � 1.2 % after 96 h of incubation with a yield of 0.189 g/l of biosurfactant. The surface tension was reduced to 46.23 mN/m � 0.46 after 72 h at pH 7.0. The presence of non-ribosomal protein sequences gave a hint of the presence of surfactin-type lipopeptide biosurfactant and Copper Translocating P-type ATPase enzyme sequences indicat-ing its presence can confer resistance against heavy metals. Strain SE I showed the potential to remove heavy metals from textile effluent viz. 67 % of Cd, 64 % of Pb, 59 % of Cu, 50 % of, Cr, and 33 % of Ni after 7 days of treatment as well exhibited decolorization of dyes ranging from 61 % � 0.15 to 95 % � 1.03, respectively. Based on the research findings, it is reasonable to conclude that surfactin biosurfactant extracted from S. sciuri subsp. rodentium strain SE I can be an alternative biosorbent and function as a bioremediation agent.
