Validation of proteomic biomarkers previously found to be differentially expressed in women with Polycystic Ovary Syndrome: a cross-sectional study

Abstract: Total haptoglobin and haptoglobin beta chain protein abundance was found to be elevated in women with PCOS compared with controls. We were unable to confirm decreased alpha-2 macroglobulin levels as reported in a previous study. Independent validation studies are required to validate early promising proteomic biomarkers in PCOS.

“…This finding supports previous proteomic studies suggesting that Hp may be a biomarker of PCOS [16,17]. In addition, a small study in the UK (n = 30 per group) using Western blotting to quantify Hp protein subunits found that total Hp and Hp beta chain protein abundance were elevated in women with PCOS compared with controls [54].…”

“…Результаты проведенного исследования не подтверждают мнения некоторых авторов [13,18,26,27] о том, что развитие СПКЯ сопряжено с признаками хронического воспаления низкой степени выраженности. Мы полагаем, что повышенные сывороточные концентрации α 2 -МГ и α 1 -АТ могут быть связаны с нарушением утилизации клетками, так как у большинства женщин поликистоз яичников ассоциируется с ожирением, что может свидетельствовать о снижении эффективности работы гепатоцитов.…”

“…Thus, the decrease in the circulating levels of hepcidin as well as α2-macroglobulin in patients with PCOS produces a synergistic effect in increasing the density of the iron exporter ferroportin in the cell surface of duodenal enterocytes and macrophages, leading to an increase in circulating levels of iron. However, a later study could not confirm the decrease in circulating levels of α2-macroglobulin in PCOS patients [ 40 ], thus underlining the need for additional studies in this area.…”
