Validation of a Commercially Available Immunoassay for the Measurement of Bovine Cardiac Troponin I

Abstract: Background: Commercially available cardiac troponin I (cTnI) assays developed for use in humans have not yet been validated for use in cattle.Hypotheses: The ADVIA Centaur TnI-Ultra immunoassay can be used for the detection of bovine cTnI. In healthy cattle, serum cTnI is undetectable or is present only in trace amounts. Methods: Purified bovine cTnI and cTnI-free bovine serum were used for the evaluation of assay performance including intra-and inter-assay precision, sensitivity, interference, linearity, and r… Show more

“…cTnI is generally considered to be a stable molecule under various storage conditions. 4,17 The reason for the increase over time in [cTnI] i in the plasma with a low [cTnI] i stored at 20-248C in this study is unknown. The [cTnI] i in the three aliquots stored at each temperature was only measured once each day.…”

“…This is consistent with results in other species, including other ruminants. 3,11,17 In one study, [cTnI] measured by the ADVIA Centaur Tn-I Ultra was 0.03 ng/ml in each of 30 healthy dairy cows; [cTnI] was 0.01 ng/ml in 23 of the cows. 17 This is unsurprising, given that other markers for loss of cellular integrity (e.g., alanine aminotransferase) are present only in low to undetectable quantities in the blood of healthy animals.…”

“…Cardiac troponin I (cTnI) is a more sensitive and persistent indicator of cardiac injury, with high tissue specificity, than other markers in the presence of marked skeletal muscle injury, liver disease, and chronic renal failure (O'Brien et al, 1997;Adin et al, 2005). The molecular structure of troponin proteins is highly conserved across species, and some current assays developed for their detection in humans have been used and validated in several other species (Oyama and Solter, 2004;Varga et al, 2009). The central hypothesis of this study is that acid-base and electrolyte disturbances associated with ARLA and secondary complications, lead to some degree of cardiovascular injury.…”

“…This presents an opportunity for field testing of large groups of cattle with rapid results that can aid in disease classification and outcome discrimination. Recently, serum cTn I has been reported for cattle having a variety of cardiogenic and non-cardiogenic disease [21][22][23][24][25][26][27].…”
