Validated LC‐MS method for simultaneous quantitation of catalpol and harpagide in rat plasma: application to a comparative pharmacokinetic study in normal and diabetic rats after oral administration of Zeng‐Ye‐Decoction

Abstract: A simple and efficient liquid chromatography-mass spectrometry (LC-MS) method was developed and validated for simultaneous quantitation of catalpol and harpagide in normal and diabetic rat plasma. Protein precipitation extraction with acetonitrile was carried out using salidroside as the internal standard (IS). The LC separation was performed on an Elite C18 column (150 × 4.6 mm, 5 µm) with the mobile phase consisting of acetonitrile and water within a runtime of 12.0 min. The analytes were detected without en… Show more

“…Additionally, the metabolism enzymes may also be found in other organs such as the kidney and gastrointestinal tract (Gardiner and Begg 2006;Smith et al, 2012). The intestinal absorption of some iridoid glycosides, like geniposide, catalpol and loganin, is enhanced by the inhibition of the intestinal P-gp activity (Chula et al, 2012;Feng et al, 2013;Li et al, 2008). Cytochrome P450 activity within the gut might have a significant effect on the bioavailability of orally administered medicines.…”

“…Although Zeng Ye Tang has been used in clinical practice since the Qing dynasty, an integrated quantity control standard has not so far been established, except for the simultaneous quantitation of catalpol and harpagide by LC–MS . In the present study, we have established an HPLC–Q‐TOF‐MS/MS method for the analysis of Zeng Ye Tang for the first time and identified 50 compounds by comparison of retention times, UV spectra, and MS spectra data (Fig.…”

“…TCM prescriptions are complex, with multiple ingredients and many potential targets. So far, only catalpol and harpagide have been detected by LC–MS analysis of plasma after treatment with Zeng Ye Tang , although other components may also be present.…”

“…Furthermore, in this study, doxorubicin was used to establish CKD rats. However, doxorubicin showed hepatotoxicity by inducing toxicity and oxidative damage in liver tissues of rats (Feng et al, 2013;Singla et al, 2014;Wided et al, 2014). The liver plays a central role in the biotransformation of xenobiotics, including drugs.…”

“…Characterization of the pharmacokinetic properties is essential for monitoring and prediction of drug disposition in vivo because of its special significance in the evaluation of drug therapeutic effect, dose adjustment and the rational use of the drug in the clinic (Feng et al, 2013). Drugs are used to treat diseases and only patients are the ultimate consumers.…”
