“…Both genes were mapped to LG08, together with the other three previously mapped chalcone synthase-like genes (chs2, chs3 and chs4). Linkage analysis established that the vps gene is linked to the three chalcone synthase-like genes as was suggested by Novak et al (2003), assuming that the vps gene is part of the same gene cluster as chs3 and chs4. However, none of the chs or vps genes mapped with our detected QTLs.…”

“…Valerophenone synthase (VPS) is involved in the first steps of biosynthesis of bitter resins and was the first chalcone synthases (CHS) homologue to be cloned (Paniego et al 1999;Okada and Ito 2001). Four other chs homologues were later isolated from hop: chs_H1, with enzymatic activity in the biosynthesis of bitter acids (Matoušek et al 2002) and chs2, chs4 and chs3, with decreased or no enzymatic activity (Novak et al 2003;Okada et al 2004). However, it has been suggested that more genes and regulatory factors are involved in the biosynthesis of alpha acids (Matoušek et al 2005).…”

“…Chalcone synthase (CHS; EC 2.3.1.74) then catalyzes the condensation of p -coumaroyl-CoA with malonyl-CoA to form naringenin chalcone (NC). A trichome-specific CHS gene, CHS_H1 , has been identified from hops ( 24 ). NC is then prenylated by HlPT1L, and further methylated by an O -methyltransferase (HlOMT1) to form XN ( 20 , 23 , 25 ).…”

“…An exon 2-specific DNA fragment (460 bp) of the VPS gene was used as a probe. Two other VPS homologous chalcone synthase genes (CHS4 and CHS_H1) are also expressed in lupulin glands (Okada et al 2004), and thus, to avoid cross hybridization, hybridization and washing were performed at 65°C (Ishii et al 1997, Novák et al 2003. NBT/BCIP (4-nitroblue tetrazolium/5-bromo-4-chloro-3-indolyl phosphate) color detection was used, with a violet-purple color indicating the hybridizing signals (closed arrowheads in Figure 2).…”
