Vaccinia viruses with mutations in the E3L gene as potential replication-competent, attenuated vaccines: Intra-nasal vaccination

Abstract: Vaccinia virus (VACV) has been used as the vaccine to protect against smallpox, and recombinant VACVs have been used to develop vaccine candidates against numerous cancers and infectious diseases. Although relatively safe for use in humans, the strains of VACV that were used as smallpox vaccines led to several complications including, progressive infection in immune compromised individuals, eczema vaccination in individuals with a history of atopic dermatitis, and encephalitis and perimyocarditis in apparently… Show more

“…Unchecked accumulation of dsRNA may also enhance the immune response through the activation of PKR and other dsRNA-sensing pathways. This approach has had success in the vaccinia virus (VACV) system, where a VACV lacking E3L (48,49) provides a useful comparison. Similar to the PKRinhibitory genes of cytomegaloviruses (26)(27)(28)(29)(30)(31)(32)(33)50), vaccinia virus E3L binds to and sequesters dsRNA and thereby prevents activation of type I interferon-induced PKR and oligoadenylate synthetase (51).…”

“…model measuring weight loss; intracranial inoculations or measures of viral titers in organs have often been performed instead. Available data indicate that the A35⌬ virus is similar in virulence to the IL-18 binding protein knockout virus (36), more attenuated than the A46R deletion mutant (42), and less attenuated than the E3L deletion, which causes a 1,000-fold attenuation (46). The A35 gene affects the development of the host adaptive immune response.…”

“…VACV mutants deleted of E3L (⌬E3L) or both K1L and C7L (⌬K1L⌬C7L) replicate abortively and express only a subset of viral genes in most mammalian cell lines (3, 24). These mutants are highly attenuated in animal hosts but are capable of eliciting immune responses, making them attractive vaccine vectors for infectious diseases and cancers (27,28). NYVAC, a VACV strain derived through deletion of 18 genes, including both K1L and C7L (27), has been used as the vector for an AIDS vaccine (2).…”
