Titanium dioxide (TiO 2 ) nanoparticles are promising biomedical agents characterized by good biocompatibility. In this study, we explored the cytotoxicity of TiO 2-x nanoparticles with a different Ti 3+ (Ti 2+ )/Ti 4+ ratio and analyzed the effeciency of eryptosis indices as a tool in nanotoxicology.Methods. Two types of TiO 2-x nanoparticles (5 nm) with various Ti 3+ (Ti 2+ )/ Ti 4+ ratios in the crystal lattice were synthesized. 1-TiO 2-x nanoparticles contained 54% Ti 4+ , 38% Ti 3+ and 8% Ti 2+ , while the relative amount of Ti 4+ and Ti 3+ in the crystal lattice of 2-TiO 2-x nanoparticles was 63% and 37%, respectively. Cell viability and cell motility induced by TiO 2-x nanoparticles were investigated on primary broblast cultures. Eryptosis modulation by the nanoparticles along with cell death mechanisms was studied on rat erythrocytes.Results. We report that both TiO 2-x nanoparticles don't decrease the viability of broblasts simultaneously stimulating cell migration. Data from in vitro studies on erythrocytes indicate that TiO 2-x nanoparticles trigger eryptosis via ROS-(1-TiO 2-x ) and Ca 2+ -mediated mechanisms (both TiO 2-x nanoparticles) suggesting that evaluation of eryptosis parameters is a more sensitive nanotoxicological approach for TiO 2-x nanoparticles than cultured broblast assays.Conclusion. TiO 2-x nanoparticles are characterized by low toxicity against broblasts, but they induce eryptosis, which is shown to be a promising tool for nanotoxicity screening. The Ti 3+ (Ti 2+ )/ Ti 4+ ratio at least partially determines the cytotoxicity mechanisms for TiO 2-x nanoparticles.

Харківський національний медичний університет, Харків, Україна Анотація. Протягом останніх двох десятиліть відмічається глобальне зростання зловживання алкоголем і частоти розладів, пов'язаних з цим. Алкоголь є провідним фактором ризику багатьох видів травм. Симптоми стану відміни алкоголю (СВА) виникають у 31% пацієнтів із травмами. У пацієнтів із політравмою та СВА, ускладненим алкогольним делірієм, відмічають запальний процес, однією з відповідей організму на цей патологічний стан є утворення активних форм кисню (АФК) нейтрофілами. Мета: визначити генерацію АФК в крові пацієнтів із політравмою та СВА з алкогольним делірієм залежно від способу седації. Об'єкт і методи дослідження. Обстежено 80 пацієнтів із політравмою середньої тяжкості та СВА, ускладненим алкогольним делірієм. Медіана віку становила 45 років. Пацієнтам 1-ї групи (n=40) в якості методу седації обрано дексмедетомідин, у 2-й групі (n=40) використовували седацію діазепамом за симптом-тригерним протоколом. Для діагностики делірію використовували метод оцінки сплутаності свідомості у пацієн тів, які перебувають у відділенні інтенсивної терапії (Confusion Assessment Method-Intensive Care Unit -CAM-ICU). Визначення АФК в еритроцитах проводили методом проточної цитометрії із застосуванням програмного забезпечення «BD FACSDiva TM » («Becton Dickinson», США). Результати. У пацієнтів із політравмою та СВА, ускладненим алкогольним делірієм, виявлено значно підвище ну генерацію АФК в еритроцитах, що зумовлює ериптоз. Висновок. Застосування дексмедетомідину може бути рекомендоване в якості седативного засобу в терапії алкогольного делірію у пацієнтів із політравмою та СВА. Ключові слова: політравма, стан відміни алкоголю, алкогольний делірій, дексмедетомідин, активні форми кисню.

Background: Semi-refined carrageenan (food additive E407a) is a widely used thickener, which has been reported to exert toxic and proinflammatory effects. In particular, there is accumulating evidence that it induces eryptosis, i.e. a programmed cell death of eryptocytes, via ROSmediated pathways. However, the role of Ca 2+ -dependent mechanisms in E407a-induced eryptosis is not elucidated.Material and methods: Semi-refined carrageenan at concentrations of 0 mg/ml, 1 mg/ml, 5 mg/ml and 10 mg/ml was incubated with blood of intact female WAG rats (n=9) for 24 h in RPMI and fetal bovine serum. After 24 h, the samples were used to obtain erythrocyte suspensions. The obtained suspensions were stained with a Ca 2+ -sensitive FLUO4 AM probe (30 min, 2.5 µM). The fluorescence of FLUO4 in erythrocytes was detected by a BD FACSCanto II flow cytometer.Results: The intracellular Ca 2+ levels are proportional to the fluorescence of FLUO4. The mean fluorescence intensities (MFI) were compared. Low levels (1 mg/ml) of E407a had no impact on Ca 2+ concentrations in erythrocytes (p>0.05). On the contrary, high concentrations (5 mg/ml and 10 mg/ml) of this food additive promoted an increase in the intracellular Ca 2+ levels. The MFI values were 2.3-and 2.5-fold higher, respectively (p<0.0001). In addition, the exposure to E407a at concentrations of 5 mg/ml and 10 mg/ml (p<0.0001) increased the percentage of cells with high FLUO4 fluorescence.Conclusion: Food additive E407a induces eryptosis in a Ca 2+dependent manner.
