UV Absorption Spectroscopic Analysis of the Molecular Orientation of a Drug Penetrated into a DPPC Membrane

Abstract: The present study quantitatively measured, via UV absorption spectroscopy, the molecular orientation of the drug chlorpromazine (CPZ) after spontaneous penetration into the gel phase of a phospholipid membrane. An L-R-dipalmitoylphosphatidylcholine (DPPC) Langmuir (L) film (a monolayer on an aqueous solution) was doped with CPZ and transferred onto a quartz substrate to form a Langmuir-Blodgett (LB) film. The transmission spectrum of the LB film was measured using a normal incident, nonpolarized UV beam. To ca… Show more

“…Via the uniaxial refractive index ellipsoid model and by comparing the theoretical approach with experimental data, previous works have established that the transmission spectrum of a Langmuir‐Blodgett film or organic thin films depends on the tilt angle (α) of the immobilized chromophores, defined with respect to the surface normal direction. In considering these findings and assuming that all PDI species are oriented in the same direction and randomly distributed in the SAM (i. e. a crude approximation), the anisotropic values of extinction coefficient ϵ || and ϵ ⊥ (i. e. parallel and perpendicular projections to the surface normal direction, respectively) of 1 could be expressed as Equation true{ϵ=32ϵSAMprefixsin2αϵ=3ϵSAMprefixcos2αwith0.166667emϵSAM:extinction0.166667emcoefficient0.166667emin0.166667emSAM …”

“…Further thermodynamic and spectroscopic studies [234,235] provided evidence for drug insertion into the hydrophobic part of the membrane, however, without modification of the chain order, which is in contrast to the action observed for the above discussed anesthetics and anti-inflammatory drugs. The experiments revealed the predominant contribution of electrostatic interactions with PO 2 -groups associated with the disruption of hydrogen bonds at the phospholipid-water interface [234].…”
