Ussing Chamber

Abstract: The Ussing chamber system is named after the Danish zoologist Hans Ussing, who invented the device in the 1950s to measure the short-circuit current as an indicator of net ion transport taking place across frog skin (Ussing and Zerahn, Acta Physiol Scand 23:110-127, 1951). Ussing chambers are increasingly being used to measure ion transport in native tissue, like gut mucosa, and in a monolayer of cells grown on permeable supports. However, the Ussing chamber system is, to date, not often applied for the inves… Show more

“…Chambers were filled with 5 mL of Krebs‐Henseleit solution (Sigma) maintained at 37°C and continuously gassed with 95% O 2 /5% CO 2 . Transepithelial resistance (TER) was monitored throughout the experiment to assess the viability of the tissue . Paracellular permeability was assessed by measuring mucosal‐to‐serosal flux of fluorescein isothiocyanate (FITC)‐labeled 4 kDa dextran (Sigma).…”

“…Dynamic in vitro methods allow the computerized control of the physiological and biomechanical conditions corresponding to each GIT compartment, taking samples from different compartments and mimicking the digestive conditions specific to infants, adults, and the elderly (Havenaar et al., 2013; Passannanti et al., 2017; Shani‐Levi et al., 2017). The cellular uptake study of biomolecules is carried out using different in vitro/ex vivo models such as: everted gut sac of the rat small intestine, to evaluate the transport of liposomes and hydrophilic molecules in different sectors of the small intestine, Ussing chambers model used for ion and other biomolecules absorption, and different cell models consisting of layers of cells obtained by in vitro growth of cells isolated from a dog kidney (Madin Darby canine kidney [MDCK] cells) or from human colon adenocarcinoma (Caco‐2 cell, HT29 cells) (Tor, 2015; Westerhout, Wortelboer, & Verhoeckx, 2015). Most studies on the permeability and toxicity of orally administered biocomponents have used Caco‐2 cells line model, since they differentiate relatively easily and form a layer of enterocyte cells with a morphology similar to the intestinal epithelial layer.…”

“…Karakteristik (232) dari Kalsium Klorida merupakan memiliki 3 struktur (73) kristal yaitu ortorombik, tetragonal, dan trigonal. Kalsium klorida boleh berfungsi sebagai sumber ion (74)(75)(76)(77)(78) kalsium dalam larutan aqua kerena sifat kalsium klorida yang boleh larut dalam air. Sifat (79) ini berguna untuk mengeluarkan ion daripada larutan (80) .…”
