Bioavailability and bioaccessibility of food bioactive compounds; overview and assessment by in vitro methods

Abstract: Oral bioavailability is the key to the bioefficiency of food bioactive ingredients; it evaluates the relationship between foods and their health benefits. The analysis of the main factors limiting the oral bioavailability (bioaccessibility, absorption, and transformation) has led to the proposal of classification systems for pharmaceuticals and nutraceuticals (Biopharmaceuticals Classification System and Nutraceutical Bioavailability Classification Scheme). Based on the relevant studies published in the last d… Show more

“…Bioaccessibility, that is, the release from the matrix, is a preliminary and important requirement for further absorption of food components. Food component bioaccessibility is not only dependent on their concentration, as food matrix composition and structure also affect the release kinetics during the digestive process [1][2][3][4]. The molecular organization of the food matrix is a determining factor since the spatial distribution of molecules, that is, the supramolecular structure, influences the order of substrate exposure to the digestive enzymes [5].…”

“…Metoprolol is commonly used as a reference [151], because its gastrointestinal membrane permeability lies in this boundary range. [153] Regarding the LogP value, it may be considered as a helpful instrument to investigate drugs' bioavailability, but effective absorption and metabolism mechanism are much more complex, and specific enzyme and sites interaction also play a relevant role. “…Bioefficiency of a nanoencapsulated material is a vital criterion to meet the metabolic needs of a consumer. Quantitative estimation of bioefficiency is usually done by determination of bioaccessibility, bioavailability and biological activity of consumed food components [ 119 ]. Bioavailability of a compound is defined as the fraction of functional component absorbed and reaching the systemic blood circulation for performing physiological functions and/or biodistribution ( Figure 16 A).…”
