In this study, we investigated the bioactive potential (antibacterial and antioxidant), anticancer activity and detailed phytochemical analysis of Selaginellarepanda (S. repanda) ethanolic crude extract for the very first time using different in vitro approaches. Furthermore, computer-aided prediction of pharmacokinetic properties and safety profile of the identified phytoconstituents were also employed in order to provide some useful insights for drug discovery. S. repanda, which is a rich source of potent natural bioactive compounds, showed promising antibacterial activity against the tested pathogenic bacteria (S. aureus, P. aeruginosa, E. coli and S. flexneri). The crude extract displayed favorable antioxidant activity against both 2,2-diphenyl-1-picrylhydrazyl (DPPH) (IC50 = 231.6 μg/mL) and H2O2 (IC50 = 288.3 μg/mL) molecules. S. repanda also showed favorable and effective anticancer activity against all three malignant cancer cells in a dose/time dependent manner. Higher activity was found against lung (A549) (IC50 = 341.1 μg/mL), followed by colon (HCT-116) (IC50 = 378.8 μg/mL) and breast (MCF-7) (IC50 = 428.3 μg/mL) cancer cells. High resolution-liquid chromatography–mass spectrometry (HR-LC–MS) data of S. repanda crude extract revealed the presence of diverse bioactive/chemical components, including fatty acids, alcohol, sugar, flavonoids, alkaloids, terpenoids, coumarins and phenolics, which can be the basis and major cause for its bioactive potential. Therefore, achieved results from this study confirmed the efficacy of S. repanda and a prospective source of naturally active biomolecules with antibacterial, antioxidant and anticancer potential. These phytocompounds alone with their favorable pharmacokinetics profile suggests good lead and efficiency of S. repanda with no toxicity risks. Finally, further in vivo experimental investigations can be promoted as probable candidates for various therapeutic functions, drug discovery and development.

Cymbopogon Proximus Essential Oil Protects Rats against Isoproterenol-Induced Cardiac Hypertrophy and Fibrosis

Cymbopogon proximus (Halfabar), a wild plant that is usually used in folk medicine due to its diuretic and antispasmodic effects, was shown to exhibit several cardiovascular protective actions such as antidiabetic, antihypertensive, antioxidant, and antinflammatory effects. However, no reports are available regarding the investigation of the role of Cymbopogon proximus essential oil on cardiac hypertrophy. Therefore, the goals of the present study were to find whether Cymbopogon proximus could alleviate isoproterenol induced‐cardiac hypertrophy and to determine the underlying molecular mechanisms. For this purpose, male albino rats were treated daily with either Cymbopogon proximus essential oil (800 μl/kg/day, ip), isoproterenol (5 mg/kg/day, ip), or the combination of both. Oil administration was started four days prior to isoproterenol administration and continued concurrently thereafter for another three days. All animal groups were euthanized 24 hours after the last dose of treatment. Thereafter, the hearts were harvested and heart weight to body weight (HW:BW) ratios were measured and sections of hearts were stained with Masson's trichrome and H&E to assess cardiac fibrosis and morphology. Our results showed that isoproterenol alone resulted in a significant increase in HW:BW ratio and marked myocardial fibrosis in treated rats. On the other hand, treatment with Cymbopogon proximus essential oil significantly reduced the isoproterenol‐mediated increase in HW:BW ratio by 55%. However, oil treatment did not affect isoproterenol mediated‐increase in myocardial fibrosis. Conclusion Cymbopogon proximus essential oil partly protects against isoproterenol‐induced cardiac hypertrophy with no effect on cardiac fibrosis. These findings provide support for the promising therapeutic effects of Cymbopogon proximus on cardiac diseases. This abstract is from the Experimental Biology 2018 Meeting. Gentianella acuta (G. acuta) has been widely used as a traditional medicine by Chinese Mongolian populations for the treatment of heart diseases and has also been tested in modern pharmacological experiments. However, the effects of G. acuta on cardiovascular damage and inflammation under conditions of hypercholesterolaemia remain unclear. The present study investigated the effects and mechanisms of the water extract of G. acuta on cardiovascular damage and inflammation caused by a high-cholesterol diet. Male Sprague-Dawley rats were fed a high-cholesterol diet for 4 weeks to establish the hypercholesterolaemia rat model, and they were administered physiological saline or 1.2 g/kg of G. acuta by gavage starting from the 15th day. After the last administration, the blood, heart and thoracic aorta samples were collected and examined. It was revealed that G. acuta treatment could ameliorate cardiomyocyte disorder and thoracic aortic vessel wall damage, reduce serum lipid levels and inflammatory factors and improve heart function. Compared with the Model group, the serum levels of triglycerides, total cholesterol, low-density lipoprotein and tumour necrosis factor-α were decreased, and the high-density lipoprotein and interleukin-10 levels were increased in the Model-G group. Moreover, in both the heart and thoracic aorta, G. acuta reduced the expression and phosphorylation of inhibitor of nuclear factor kappa-B kinase β (IKKβ), inhibitor of NF-κB-α (IκBα) and p-nuclear factor kappa-B (NF-κB). Therefore, G. acuta may exert an inhibitory effect on the IKK/IκB/NF-κB signalling pathway to protect the heart and thoracic aorta in hypercholesterolaemic rats.
