Use of doubly protonated molecules in the analysis of cathedulins in crude extracts of khat (Catha edulis) by liquid chromatography/serial mass spectrometry

Abstract: Analysis of crude methanolic extracts of fresh khat (Catha edulis) by liquid chromatography/mass spectrometry (LC/MS) revealed the presence of 62 cathedulin alkaloids (compared with 15 published structures). Many cathedulins generated doubly protonated molecules following electrospray ionisation and the ratio of doubly to singly protonated species could be manipulated by adjusting the electrospray capillary position and source conditions. By selecting the doubly protonated species for serial mass spectrometric… Show more

“…Cathedulins, for example, comprise one group of alkaloids with up to 62 members. 15,16 Khat also contains vitamin C and tannins as well as small amounts of essential oils, sterols, triterpenes, thiamine, riboflavin, niacin, iron, and amino acids. 15,17 Despite the extensive literature about Khat chewing, information concerning its effects on oral health is inadequate.…”

“…Overall, it seems that khat is less appealing to women, although in Somalia chewing has recently become more popular among middle-class and educated women. Cathinone is unstable and undergoes decomposition reactions after harvesting and during drying or extraction of the plant material (8,(24)(25)(26). Decomposition leads to a 'dimer' (3,6-dimethyl-2,5-diphenylpyrazine) and possibly to smaller fragments.…”

“…Khat contains a number of pharmacologically active compounds (Kite et al, 2003). Cathinone is a major alkaloid component in fresh Catha leaves, but is relatively unstable and is rapidly metabolised into norpseudoephedrine (cathine) and norephedrine.…”
