Urm1: an essential regulator of JNK signaling and oxidative stress in Drosophila melanogaster

Abstract: Ubiquitin-related modifier 1 (Urm1) is a ubiquitin-like molecule (UBL) with the dual capacity to act both as a sulphur carrier and posttranslational protein modifier. Here we characterize the Drosophila melanogaster homologues of Urm1 (CG33276) and its E1 activating enzyme Uba4 (CG13090), and show that they function together to induce protein urmylation in vivo. Urm1 conjugation to target proteins in general, and to the evolutionary conserved substrate Peroxiredoxin 5 (Prx5) specifically, is dependent on Uba4.… Show more

“…This result contrasts with some studies showing that JNK activity is necessary for UVR-induced apoptosis145960. However, our results are consistent with studies suggesting that JNK is also a key modulator of oxidative stress-mediated cell survival14606162. Discrepancies between studies may be explained by the use of different UV sources: whereas a UVA/B source that reproduces the UVR solar spectrum was used in our study, cells were exposed to UVC in the aforementioned studies145960.…”

“…Both roles are exchangeable among eukaryotes 1011 and based on dual-function Urm1-like proteins in archaea and bacteria 1213, conserved Urm1 systems appear to be important in all domains of life. In line with this, their inactivation triggers stress-induced growth defects in microbes, organ underdevelopment in plants and, strikingly, lethality in flies 348121314151617.…”

“…Probably, this may help to adapt to conditions known to be URM1 pathway related, i.e. ROS, thermo-stress, S-depletion 6111738394041. However, with attachment of ubiquitin-like modifiers such as SUMO regulating the activity of their targets in ways different from Urm1 2484950, how precisely urmylation impacts on its targets is still not clear.…”

“…A role of Urm1 in oxidative stress has strongly been supported by the observation of increased levels of urmylation and altered sensitivity during oxidative stress in multiple model systems, including yeast, mammalian cells and in D . melanogaster [ 17 , 19 , 29 ]. However, the attempts to identify Urm1 targets and understand the biological role of target protein urmylation during these processes have been sparse.…”
