Urethral striated muscle and extracellular matrix morphological characteristics among mildly diabetic pregnant rats: translational approach

Abstract: Mild DM and pregnancy can lead to a time-dependent disorder and tissue remodeling in which the urethral striated muscle and ECM has a fundamental function.

“…The STZ model generally represents the etiology of type 1 diabetes or advanced stages of type 2 diabetes (Bell Jr. and Hye, 1983). The STZ model has been extensively studied and used to assess the complications of type 1 diabetes (Badalzadeh et al, 2015;Piculo et al, 2014), learning and memory (Bellush and Rowland, 1989;Flood et al, 1990), natural hedonic processing and drug reward (Carr, 1994;Carr et al, 2000;Galici et al, 2003b;O'Dell et al, 2014). Thus, the STZ model represents a common method of inducing diabetes via disruptions in insulin signaling.…”

“…At term pregnancy, the morphological analysis of the extracellular matrix and urethral striated muscle from urethrovaginal tissues showed that the urethral layers were thin, disorganized, and atrophic and there were more fibrosis replacement and collagen fiber deposition associated to muscle atrophy. In addition to structural analyses, one of the most significant findings was the colocalization of fast and slow fibers and a steady decrease in the proportion of fast to slow fibers by immunohistochemical examination in the mild diabetic pregnant rats [64]. Diabetes is related to accumulation of reactive oxygen species, and tissue ischemia which can interactively or independently contribute to the myopathy causes of skeletal muscle dysfunctions [65, 66].…”

“…Glycogen granules, lipid droplets, and numerous mitochondria were apparent in the striated muscle cells [64]. Lipid droplets are not membrane bound, and their number and size may vary considerably among different muscle types.…”
