Urethral seam formation and hypospadias

Abstract: Knowledge of the formation of the normal male urethra may elucidate the etiology of hypospadias. We describe urethral formation in the mouse, show the similarities and relevance to human urethral development, and introduce the concept of the epithelial seam formation and remodeling during urethral formation. Three mechanisms may account for epithelial seam formation: (1) epithelial-mesenchymal transformation similar to that described in the fusion of the palatal shelves, (2) apoptosis, and/or (3) tissue remode… Show more

“…Later, a UF develops further toward the midline and finally fuses in the midline to generate the tubular penile urethra 1 and the progressive formation of the UF leading to midline fusion from proximal to distal regions of the GT has been reported. 4 Within the GT, we have identified a transient epithelial structure located at the distal end of UP termed distal urethral epithelia (DUE). 5,6 The DUE expresses several growth factors, including Fgf (fibroblast growth factor), Bmps (bone morphogenetic proteins) and Sonic hedgehog (Shh).…”

“…Because of the presence of TUNEL-negative stratified epithelium (e.g., Figs. 7 and 8 in Baskin et al 2001), the usual type of apoptosis with DNA fragmentation seemed unlikely to occur in the present sc9758-positive sites. Therefore, we hypoth- esized that sc9758 cross-reacted with epithelium-related substances produced by the death process.…”

“…The preputial lamella also undergoes luminization to free the prepuce from the glans of the penis or clitoris. Notably, according to figures presented by Baskin et al (2001), the TUNEL method did not demonstrate any apoptotic cells in the preputial lamella of fetal mice. An epithelial plug has also been reported in the human fetal anal orifice (Nievelstein et al 1998).…”

“…Failed seam formation during fusion of the urethral folds between fetal weeks 8 and 16 result in hypospadias. 7 Any gene having a role during the development of urogenital organs in the critical period is therefore a candidate gene for hypospadias. Mutations in the MID1 gene cause the X-linked form of Opitz G/BBB syndrome (OS, characterized by midline abnormalities such as hypertelorism, cleft lip/palate, tracheo-esophageal abnormalities, cardiac defects and hypospadias).…”
