Upregulation of Vascular Endothelial Growth Factor Expression in the Kidney Could Be Reversed Following Treadmill Exercise Training in Type I Diabetic Rats

Abstract: Background: Nephropathy is a significant complication of diabetes mellitus, which is associated with high morbidity and mortality. Exercise training has been shown to have renoprotective effects in diabetes. Unregulated vascular endothelial growth factor (VEGF) has been demonstrated in the diabetic kidney. Thus, the aim of our study is to illustrate the impact of endurance exercise training on the renal VEGF expression in type I diabetic rats.

“…Furthermore, moderate-intensity exercise has been reported to reduce advanced glycation end products and ameliorate early diabetic nephropathy in obese Zucker rats 35 . Therefore, we agree with the conclusion that exercise training may reduce VEGF upregulation in diabetic kidneys by reducing hyperglycemia and increasing insulin sensitivity 34 . Similar favorable alterations occurred in kidney tissue in response to exercise performed at various frequencies.…”

“…VEGF upregulation has been reported in diabetic kidney tissue similar to our study 5 . In a study conducted by Al-Jarrah et al 34 rats were given treadmill exercise at a speed of 18 m/min 40 min/day for 5 days/week and diabetes-related elevation of VEGF levels declined. In our study, VEGF expression declined in the exercise groups, in line with the literature.…”

“…Immunohistochemistry of IL-1β, TNF-α, and NF kappa B p65 was performed using a previously described protocol [25,2832]. Briefly, deparaffinization and rehydration were performed in the prepared 4-μm-thick paraffin-embedded gastrocnemius skeletal muscle sections.…”

“…A digital camera was used to photograph 10 random regions of each section that were subsequently examined for IL-1β expression, TNF-α expression, and nuclear localization of NF kappa B in the gastrocnemius skeletal muscle. Then, total pixels region of positive staining (Figures 1C, 2C, 3C) was computed in proportion to the entire pixels region in each photographed area of each section using Adobe Photoshop software, as described previously [25,2832]. After that, the mean of the pixels region of positive immunostaining relative to the entire pixels region was computed for every animal in the 2 different groups.…”

“…Then, serial longitudinal 4-μm–thick paraffin-embedded sections were cut. The sections were processed via immunohistochemistry according to previously described protocols [ 28 , 30 36 ]. To summarize, the sections were firstly deparaffinized and rehydrated.…”
