Untargeted metabolomics and transcriptomics identified glutathione metabolism disturbance and PCS and TMAO as potential biomarkers for ER stress in lung

Abstract: Endoplasmic reticulum (ER) stress is a cellular state that results from the overload of unfolded/misfolded protein in the ER that, if not resolved properly, can lead to cell death. Both acute lung infections and chronic lung diseases have been found related to ER stress. Yet no study has been presented integrating metabolomic and transcriptomic data from total lung in interpreting the pathogenic state of ER stress. Total mouse lungs were used to perform LC–MS and RNA sequencing in relevance to ER stress. Untar… Show more

“…Among the significantly influenced pathways, glutathione metabolism was closely related to OS and ER stress. 52,53 The increased GPX3 expression in the enriched glutathione metabolism pathway was in line with the elevated hepatic GSH-Px concentration obtained by biochemical analyses in the current study. Moreover, the expression of ACOX1 in the peroxisome pathway was decreased by DEX treatment.…”

“…Metabolomic techniques such as targeted Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) are used for the quantification of specific metabolites like glutathione, directly involved in redox homeostasis [ 82 ]. Untargeted Metabolomics gives a comprehensive overview of metabolic changes due to oxidative stress and can provide potential biomarkers for cancer [ 83 ].…”
