Unravelling the Novel Effects of Three Volatile Compounds in Preventing Fibril Formation of Disease Related Tau and α-Synuclein Proteins- Towards Identifying Candidate Aromatic Substances for Treating Neurodegenerative Diseases

Abstract: Background: The aggregation of tau and α-synuclein into fibrillary assemblies in nerve cells is the molecular hallmark of Alzheimer’s and Parkinson’s diseases, respectively. In our previous studies, we investigated the anti-amyloidogenic effects of three different aroma-producing (volatile) compounds including cinnamaldehyde, phenyl ethyl alcohol, and TEMED on the fibrillation process of HEWL, as a model protein. Our previous results showed that while TEMED was able to completely stop the process of fibril for… Show more

“…The pellet was dissolved in 20 mM Tris–HCl (pH 8), sterile-filtered through a 0.22 µm syringe filter, and loaded onto a 5-ml HiTrap Q FF anion exchange chromatography column (GE Healthcare). α-syn was eluted at 300 mM NaCl and the pure protein (confirmed by SDS-PAGE electrophoresis) 65 was dialyzed overnight against phosphate-buffered saline (PBS) (137 mM NaCl, 2.7 mM KCl, 10 mM Na 2 HPO 4 , 1.76 mM KH 2 PO 4 , pH 7.4). The protein concentration was determined by measuring the absorbance at 275 nm 64 .…”
