Undetectable Free Aromatic Amino Acids in Nails of Breast Carcinoma: Biomarker Discovery by a Novel Metabolite Purification VTGE System

Abstract: Conclusion: This paper provides a proof of concept for a novel and specifically developed VTGE process that showed first evidence on the undetectable level of FAAAs in nails of breast cancer patients as metabolite biomarkers. Here, the authors propose the potential use of a VTGE-assisted process to achieve metabolomic discovery in nails of breast cancer and other tumor types.

“…The primary goals were to study the nail lysate in the context of nicotine and nicotine-derived metabolites with the assistance of a novel vertical tube gel electrophoresis (VTGE) metabolite purification system and identification of these metabolites with the help of LC-HRMS (Sharma et al, 2019; Mitruka et al, 2020). A working model of novel VTGE system is presented in Figure S1.…”

“…The authors agree that reported novel approach on identification of nail metabolites also detects other routinely used drugs and metabolic products as shown in the TIC of healthy and OSCC subjects. Our lab and existing literature have attempted to address the accumulation of various types of drugs, metabolized drugs, environmental toxins (Oyoo-Okoth et al, 2010; Krumbiegel et al, 2016; Mitruka et al, 2020; Sharma et al, 2020). However, this finding presents a first report on the detection of oncometabolites such as 1-MNA, nicotine imine and N-methylnicotinium ion and their possible link with the metabolic-epigenetic signaling axis.…”

“…In the case of nails, even post-mortem specimens can be successfully collected and investigated [72]. According to the cited study [36], nails are a promising matrix for evaluating cancer-related alterations in amino acid metabolism in patients.…”

“…Kynurenine, which is a metabolized product of tryptophan, may promote differentiation of T regulatory cells, which might help in cancer-mediated evasion of the immune system [94]. The decreased levels of phenylalanine mentioned in a few studies [36,40,46] might be caused by the increased demand for this amino acid by cancer cells [95]. As proline can be used as an energy source and as a precursor for protein synthesis, the need for this amino acid is increased and thus, proline biosynthesis is increased.…”
