Understanding the Stony Bridge between Osteoporosis and Vascular Calcification: Impact of the FGF23/Klotho axis

Abstract: A relationship between osteoporosis (OP) and vascular calcification (VC) is now proposed. There are common mechanisms underlying the regulation of them. Fibroblast growth factor- (FGF-) 23 and Klotho are hormones associated with the metabolic axis of osteovascular metabolism. Most recently, it was suggested that the FGF23-klotho axis is associated with increasing incidence of fractures and is potentially involved in the progression of the aortic-brachial stiffness ratio. Herein, we discussed the potential role… Show more

“…Fibroblast growth factor 23 (FGF23), a phosphatide hormone secreted by osteocytes and osteoblasts, inhibits the activation of VD and induces excretion of Pi through proximal renal tubular epithelial cells [ 12 ]. Excessive action of FGF23 will impair bone mineralization, leading to hypophosphatemic rickets/osteomalacia.…”

“…FGF23 is mainly produced by the skeletal system, which is different from FGF2 and FGF18 production. FGF23 and its receptor Klotho protein, participate in the aging process by regulating phosphate homeostasis, vitamin D metabolism, Wnt, and the p53/p21 signalling pathway, which are involved in the pathophysiology of osteoporosis and vascular calcification ( 97 ).…”

Historisch gewachsen und aufgrund der Mineralisation von Geweben diskutieren mehrere Arbeiten einen Zusammenhang zwischen Osteopathologien und dem Prozess der arteriosklerotischen Kalzifikation [4][5][6].…”
