Ultrasensitive and Simultaneous SERS Detection of Multiplex MicroRNA Using Fractal Gold Nanotags for Early Diagnosis and Prognosis of Hepatocellular Carcinoma

Abstract: Sensitive and simultaneous detection of multiple cancer-related biomarkers in serum is essential for diagnosis, therapy, prognosis, and staging of cancer. Herein, we proposed a magnetically assisted sandwich-type surface-enhanced Raman scattering (SERS)-based biosensor for ultrasensitive and multiplex detection of three hepatocellular carcinoma-related microRNA (miRNA) biomarkers. The biosensor consists of an SERS tag (probe DNA-conjugated DNA-engineered fractal gold nanoparticles, F-AuNPs) and a magnetic capt… Show more

“…Wu et al have displayed the ultrasensitive and simultaneous SERS detection of multiple miRNAs using fractal gold-nanotags labeled by reporters. [22] Cheng et al have exhibited the SERS detection of free proteins in serum, showing that the simultaneous assay of free and total prostate-specific antigen provides better precision for the diagnosis of prostate cancer. [23] The latest research has shown that SERS is used to specifically characterize exosomes.…”

“…The multiplex detection of three hepatocellular carcinoma-related miRNA (miRNA-122, miRNA-21, and miRNA-223) biomarkers using magnetically assisted sandwich-type SERS was recently demonstrated [ 68 ]. The detection is based on Raman dye-modified fractal AuNPs and Ag-coated magnetic NPs.…”

“…Surface-enhanced Raman scattering (SERS) is a developing analytic technology that can amplify greatly the Raman spectral signals of analytes absorbed on some special substrate materials. , Because of the advantages, including quick response, rich fingerprint information, high sensitivity, wide detection range, and simple pretreatment, SERS analysis shows great application potential in many fields such as biological tissue, , food safety, environmental detection, , drug analysis, and other fields. However, there is still a certain distance for this novel sensing technology to wide spread, due to the limitations of the SERS substrate. In general, most of the current SERS substrates suffer from the following problems: (1) it is difficult to take into account both high sensitivity and long-term stability. ,, Fresh Ag nanostructures usually show much better SERS activities than Au nanomaterials. ,,, (2) The current SERS substrates are either in solid state or liquid state. , Usually, solid-state SERS substrates are much more popular than liquid-state SERS substrates because they are easier to operate.…”
