Ultrahigh‐pressure dual online solid phase extraction/capillary reverse‐phase liquid chromatography/tandem mass spectrometry (DO‐SPE/cRPLC/MS/MS): A versatile separation platform for high‐throughput and highly sensitive proteomic analyses

Abstract: Capillary RPLC/ESI-MS (cRPLC/ESI-MS) is one of the most powerful analytical tools for current proteomic research. The development of cRPLC techniques coupled online to a mass spectrometer has focused on increasing the separation efficiency, detection sensitivity, and throughput. Recently, the use of high-pressure (over 10,000 psi) LC systems that utilize long, small inner diameter capillary columns has gained much attention for proteomic analyses. In this study, we developed an ultrahigh-pressure dual online S… Show more

“…We have incorporated the FOLDS into a DO-SPE/RPLC system, which has been reported previously (28). Briefly, this system incorporated the following fluidic components: a six-port injection valve with a 5-l sample loop, three four-port valves, and two six-port valves (VICI Valco, Houston, TX) rated to 15,000 p.s.i.…”

“…LC-MS/MS Analysis-The peptide samples were separated using a homebuilt ultrahigh-pressure dual on-line solid phase extraction/capillary reverse-phase liquid chromatography system, having a maximum operating pressure of 10,000 psi, as previously described (32). Briefly, the system is equipped with two reverse-phase liquid chromatography capillary columns (75 m ID ϫ 360 m OD ϫ 80 cm length, C18-bonded particles, 3 m, 300 Å pore size, Jupiter) (Phenomenex, Torrance, CA), which were manufactured in-house through slurry packing.…”

“…LC/MS/MS Experiments-The tryptic yeast peptides were separated using a modified version of our ultrahigh pressure dual on-line solid phase extraction/capillary reverse-phase liquid chromatography (DO-SPE/cRPLC) system, with a maximum operating pressure of 10,000 p.s.i., that is described in detail elsewhere (13). Briefly we incorporated SCX separation (operated using an Agilent 1100 nano-LC system) on line with the DO-SPE/cRPLC system (operated using two ISCO pumps, 100 DM) to construct an on-line two-dimensional LC system.…”
