Über aktivierte Ester. III. Umsetzungen aktivierter Ester von Aminosäure‐ und Peptid‐Derivaten mit Aminen und Aminosäureestern

Abstract

Volumcn XXXVIII, E'asciculus I (1 955)h-0. 9-10. 83 erhitzt; dabei schied sich langsam Triathylamin-hydrochlorid aus. Nach dem Erkalten wurde in der ublichen Weise aufgearbeitet. Aus dem alkalischen Auszuge konnten 0,3 g (7%) Carbobenzoxyglycin durch Ansauern zuruckgewonnen werden. Der Neutralteil war ein 01 (5,2 g), welches aus Ather-Petrolather kristallisiert wurde: 4,1 g (83%) farblose Kristalle (Smp. 62-63O) von Carbobenzoxy-glycin-cyanmethylester. Zur Analyae wurde eine Probe zweimal aus Ather umkristallisiert ; der Smp. blieb unverandert 69 -7OO. Carbobenzoxyglycin-cyanmethylester ( I ) , M e t h o d e 11: 20,9 g (0,l Mol) Carbobenzoxyglycin wurden mit 21 ml (0,15 Mol) Triathylamin ubergossen, wobei unter Erwarmung eine viskose Masse entstand. Zugabe von 19,l ml (0,3 Mol) Chloracetonitril liess eine Losung eiitstehen, welche sich spontan auf 50-60° erwarmte. Die Mischung wurde '/z Std. auf 70° envarmt, wobei sie sich gelb farbte und etwas Triathylaminhydrochlorid ausschied. Nach ublicher Aufarbeitung wurde ein fast farbloses, kristallisierendes 01 erhalten, welches aus Ather 23,4 g (94%) Carbobenzoxyglycin-cyanmethylester in farblosen, tafelformigen Kristallen vom Smp. 69-70° lieferte. Misch-Smp. mit dern Analysenpraparate (Smp. 69 -7OO) ohne Depression. Die Elementaranalysen wurden in unserer mikroanalytischen Abteilung unter drr Lcitung von Herrn Dr. H . Gysel ausgefuhrt. SUMMARY.A number of activated esters of amino acid and peptide derivatives were reacted with various organic amines, including amino acid esters. The good yields obtained, and the mild reaction conditions make it seem possible that the activated ester method of acylation mag be of good use in peptide synthesis. Aminolysis by aromatic amines and by secondary amines proceeds only slowly, but the reaction can be greatly facilitated t,hrough the catalytic action of strong tertiary amines and of acids.
