Melaleuca leucadendron L. is a plant whose almost all parts (bark, leaves, twigs, and fruit) can be used as medicine, such as antioxidants, antifungals, sedative effects, and anti-hyaluronidase. This research was conducted to determine tyrosinase inhibition activity and compound content of M. leucadendron leaves. Maceration of M. leucadendron leaves was done in methanol, then carried out liquid-liquid fractionation with n-butanol, chloroform, and water. Methanol extract, butanol fraction, chloroform fraction, and water fraction were tested for phytochemical screening and tyrosinase inhibition using L-DOPA substrate with an ELISA plate well reader. The results of the tyrosinase inhibition activity test at concentrations of 100, 1000 and 10000 μg/mL respectively showed that methanol extract 29.532%, 55.227%, 89.583%; butanol fraction 29.313%, 59.174%, 94.737%, chloroform fraction 21.820%, 24.671%; 53.765%; water fraction 24,086%, 47.661%, 91.118%. Inhibition of the tyrosinase enzyme is shown through the IC50 value from methanol extract, butanol fraction and water fraction, and kojic acid as a positive control, respectively 645.438 μg/mL, 517.935 μg/mL, 669.403 μg/mL, 50.064 μg/mL. Phytochemical screening showed that the extract and fraction contained tannins, flavonoids, saponins, terpenes, and steroids. These results indicate that the butanol fraction is more potent as an anti-tyrosinase agent than the others.

Characteristic of Seaweed Porridge Eucheuma cottonii and Turbinaria conoides as Raw Peel off Mask

Peel off seaweed masks have active ingredients and antioxidants are needed. The use of seaweed porridge as raw material for peel off masks is an alternative because it zero waste and low production costs. The purpose of this study was to determine the characteristic and the best ratio porridge seaweed of Eucheuma cottonii and Turbinaria conoides as a raw material for peel masks, and also peel masks from the ratio of the best seaweed porridge. The porridge seaweed E. cottonii and T. conoides were characterized including moisture, pH, phytochemicals and antioxidants. The ratio of the best seaweed pulp is formulated for peel off skin masks that characterized by pH, viscosity, dry time, dispersion, antioxidant, and sensory. The results showed that the best porridge ratio of E. cottonii and T. conoides was obtained at a ratio of 1:1 with a pH value of 6.3; IC50 value is 137.36 ppm. Porridge E. cottonii and T. conoides contain a bioactive components. Peeled masks with a 1:1 ratio of seaweed porridge formula have the characteristics of drying time for 25.50 minutes, dispersing power of 5.5 cm, viscosity of 7,200 cP, pH value 6.81, and containing antioxidants 902.58 ppm. Sensory peel off masks seaweed formulas against appearance, color, odor, and texture are no different from peel masks without seaweed formulas and commercial masks.

Tyrosinase is an enzyme that plays a role in the formation of melanin on two reactions, namely monophenolase and diphenolase. The process of melanin formation can be reduced through the inhibition mechanism of the tyrosinase enzyme. Seaweeds are natural organisms that have recognized lightening agents. Sargassum plagyophyllum contains secondary metabolite compounds such as alkaloids, steroids, flavonoids, saponins, and tannins while Eucheuma cottonii contains alkaloids and terpenoids. The aim of this research was to determine the tyrosinase inhibitory activity of seaweeds. The yield of methanol extract amounted to 1.342% in S. plagyophyllum and 0.2599% in E. cottonii. The inhibitory activity of tyrosine is performed by measuring enzymatic reaction using L-tyrosine (monophenolase) and L-DOPA (diphenolase) substrates and presented as IC50 value. The IC50 values of kojic acid as a positive control were 15.566 μg/mL for monophenolase and 29.156 μg/mL for diphenolase. The IC50 values of S. plagyophyllum and E. cottonii methanol extracts for monophenolase were 2195.206 μg/mL and 2691.478 μg/mL, respectively. The IC50 value of methanol extract was 1769.336 μg/mL for S. plagyophyllum and 2631.648 μg/mL for E. cottonii on diphenolase. Methanol extracts from these two species of seaweed have the potential to be developed as bioactive compounds in skin lightening cosmetics.
