Two subpopulations of human triple-negative thymic cells are susceptible to infection by human immunodeficiency virus type 1 in vitro

Abstract: Some infants infected with human immunodeficiency virus type 1 (HIV-1) rapidly develop a fatal disease characterized by a severe lymphopenia. To explain the immune dysfunction, we proposed a mechanism by which a nongeneration of CD4+ T cells is caused by HIV-1 infection of thymic cells. To examine this hypothesis, we infected primary triple-negative (TN; phenotypically CD3- CD4- CD8-), CD1a- TN, or CD1a+ TN thymic cell subsets. Our data indicate that by flow cytometry, TN, CD1a- TN, and CD1a+ TN cells remain C… Show more

“…The complex pattern of primary-receptor and coreceptor expression is probably reflected by an equally complex pattern of HIV/SIV infection in vivo. Many of the stromal and lymphoid cells in the bone marrow, thymus, lymph nodes, and spleen are infected by HIV/SIV (2,6,21,29,53). In the early stages of HIV/SIV infection of lymph nodes, the majority of viral replication occurs primarily in macrophages and lymphocytes, whereas in the later stages, viral RNA is concentrated primarily in the germinal center, colocalizing with the follicular dendritic cells (FDC) (22,44).…”

“…HIV-1 infection of non-CD4-bearing cells has been shown to occur in implants in the SCID-hu mouse and in vitro in cultured thymocytes (2,39,44). Others have reported HIV-1 infection of CD4-negative thymocytes, but these studies did not rule out involvement of a CD4-mediated pathway in infection (15,47). HIV-1 has also been found harbored in CD8 SP lymphocytes in vitro in various culture systems and in the peripheral blood and lungs of patients with AIDS (11,27,31,38).…”

“…These culture conditions form the basis of the slow/low versus rapid/high biological phenotype of CCR5 and CXCR4 tropic primary isolates in PBMC (5). In both PBMC and the SCID-hu mouse, the distribution of thymocyte coreceptors described in this study is a major determinant of the biological phenotype of NL4-3 and JR-CSF (27,28,64,71,74). The expression of CXCR4 on the immature CD3 Ϫ /CD4 ϩ/low / CD8 ϩ thymocytes may lead to a rapid productive infection and destruction of this actively proliferating cell population.…”
