Two-Step Mechanism of Cellular Uptake of Cationic Gold Nanoparticles Modified by (16-Mercaptohexadecyl)trimethylammonium Bromide

Abstract: Cationic colloidal gold nanorods (GNRs) have a great potential as a theranostic tool for diverse medical applications. GNRs' properties such as cellular internalization and stability are determined by physicochemical characteristics of their surface coating. GNRs modified by (16-mercaptohexadecyl)trimethylammonium bromide (MTAB), GNRs, show excellent cellular uptake. Despite their promise for biomedicine, however, relatively little is known about the cellular pathways that facilitate the uptake of GNRs, their … Show more

“…As expected, the POSAB GNRs aggregated immediately after dilution in PBS and Dulbecco Modified Eagle's medium (DMEM) as was observed by the change in absorption spectra where extensive broadening and decrease of intensity of the longitudinal peak was observed. However, in contrast to MTAB GNRs, POSAB GNRs tend to form aggregates also in DMEM with 10% fetal bovine serum (10% FBS/DMEM) (Figure A,B) despite the presence of serum proteins that previously showed a nanoparticle anti‐aggregation effect . To conclude these data, the surface modification of GNRs by POSAB allowed successful replacement of the CTAB from GNRs surface, however, provided less but reasonable stable GNRs compared to highly stable alkanethiol‐(MTAB)‐covered GNRs .…”

“…Furthermore, we found that POSAB GNRs distributed subcellularly were forming aggregates in the cytoplasm close to the cell nucleus like MTAB GNRs. We performed a colocalization assay with POSAB GNRs and lysosomal marker LAMP‐1 to verify its lysosomal destination (Figure E), which was observed for MTAB GNRs . Figure F shows a quantitative analysis of MTAB GNRs and POSAB GNRs colocalization with LAMP‐1 expressed as thresholded Manders' coefficient (tM).…”
