Two sources of dopamine for the hippocampus

Abstract: Dopaminergic signalling is established as playing an important role in novelty related modulation of hippocampal memory. Two recent studies have identified the noradrenergic fibres originating in the locus coeruleus as an additional source of neurotransmitter acting on dopaminergic receptors in the hippocampus.

“…Novel genetic and optogenetic techniques make it possible to study the function of neuromodulator nuclei with greater specificity (Carter et al, 2010;Eban-Rothschild et al, 2016). Using these methods, recent studies dissected VTA and LC projections in hippocampus and found that dopamine release from dense LC projections is the primary cause of learning and memory in certain tasks (Kempadoo et al, 2016;Takeuchi et al, 2016;McNamara and Dupret, 2017). Optogenetic stimulation of LC terminals in hippocampus enhanced DA together with NA and specific LC lesion reduced both neurochemicals (Kempadoo et al, 2016).…”

“…Dopaminergic projections from both the substantia nigra pars compacta and the ventral tegmental area are important for memory processing. 12,13 Also, activation of cholinergic projections activation from the medial septum/diagonal band is sufficient to induce 40-Hz network oscillations in the hippocampus in vitro, thus playing an important role in hippocampal memory processing. 14 Serotonergic projections from the median raphe nucleus to the ventral hippocampus have also been consistently described 15 and are involved in the retrieval of fear memories.…”

“…In particular, via its action on ß-adrenoceptors in the hippocampus, the LC modulates long-term potentiation (LTP) and long term-depression (LTD), key determinants of synaptic plasticity and memory 29,30 . Recent optogenetic work has confirmed a causal role of LC activity in hippocampal LTP and memory enhancement, potentially via co-release of dopamine [31][32][33] .…”
