Two new vinblastine-type N-oxide alkaloids from Catharanthus roseus

Abstract: Two new vinblastine-type N-oxide alkaloids, 17-desacetoxyvinblastine N'b-oxide (1) and 20'-deoxyvinblastine N'b-oxide (2), were isolated from the leaves of Catharanthus roseus. The structures of 1 and 2 were established by the analysis of their nuclear magnetic resonance and HR-ESI-MS spectroscopic data. All alkaloids were evaluated for their cytotoxic activities against the human hepatocellular carcinoma (HepG2) cell line, human colorectal carcinoma (Lovo) cell line and human breast carcinoma (MCF-7) cell lin… Show more

“…This is one of the most precious anticancer plants indigenous to Madagascar. Previously, approximately 30 bis-indole alkaloids and over 60 monomeric indole alkaloids have been isolated from the aerial parts and roots of C. roseus [ 85 , 86 ]. Wang et al [ 87 ] isolated three new cytotoxic dimeric indole alkaloids ( 32 – 34 ) along with other five known compounds from the whole plant of C. roseus collected from China ( Table 4 ).…”

“…Therefore, the employment of phytochemicals as complementary or first-line treatments for cancer came as a recurring alternative over the years [7,8]. Actually, different plantderived medicines are currently employed in chemotherapy medication for different cancer treatments, such as the alkaloids vincristine and vinblastine, originally obtained from Catharanthus roseus, being used in the treatment of acute lymphocytic leukemia, acute myeloid leukemia, Hodgkin's disease, neuroblastoma, and small cell lung cancer, bladder cancer, brain cancer, and testicular cancer, among others [9]. Paclitaxel, obtained from Taxus brevifolia, is used to treat breast, ovarian, lung, bladder, prostate, as well as other types of solid tumor cancers [10].…”

“…Most of the alkaloids isolated in the previous years from this plant were reported by Heijden et al, (2004) [194] . With the introduction of the HR-EI-MS technique, a greater number of new alkaloids have been reported in the past few years (Wang et al, 2011;Zhang et al, 2013) [206,116] . Recently, Zhang et al, (2013) [116] [204] isolated Catharoseumine from the whole plant at the concentration of 0.786 mg/kg.14', 15'didehydrocyclovinblastine, 17-deacetoxyvinamidine and 17deacetoxycyclovinblastine was also isolated by Wang et al…”
