Twenty proteins containing a C-terminal SOCS box form five structural classes

Abstract: The four members of the recently identified suppressor of cytokines signaling family (SOCS-1, SOCS-2, SOCS-3, and CIS, where CIS is cytokine-inducible SH2-containing protein) appear, by various means, to negatively regulate cytokine signal transduction. Structurally, the SOCS proteins are composed of an N-terminal region of variable length and amino acid composition, a central SH2 domain, and a previously unrecognized C-terminal motif that we have called the SOCS box. By using the SOCS box amino acid sequence … Show more

“…We have begun to examine transcript levels for several known regulators of the IFN-g response in control and WT cells following treatment with IFN-g. These have included IRF-1, IRF-2, ISGF3-g-p48, and SOCS1 and SOCS3, two negative regulators of cytokine action (Hilton et al, 1998;Nguyen et al, 1997). To date, the only di erences detected between these two cell lines has been in the basal levels of ISGF3g-p48 which were expressed approximately threefold higher in WT cells as previously reported (Weihua et al, 1997).…”

“…So far, eight members of this family of proteins have been identi®ed (Hilton et al, 1998;Kovanen and Leonard, 1999). Expression of CIS is regulated at least in part by cytokine-induced STAT proteins (Matsumoto et al, 1997;Verdier et al, 1998).…”

“…Like patched, Hip is transcriptionally activated by hedgehog signalling, and it has also been proposed to be a negative regulator of signalling (Chuang and McMahon, 1999). Another gene shown to be responsive to hedgehog signalling in both the somites and limb buds is a member of a novel family of SOCS (suppressor of cytokine signalling) box-containing WDproteins and has been termed both WSB1 and SWIP-1 (Hilton et al, 1998;Vasiliauskas et al, 1999). This gene is one of the earliest markers to respond to sonic hedgehog in the limb, but its role in the signalling pathway remains unclear (Vasiliauskas et al, 1999).…”
