Turning up the Heat with Therapeutic Epigenome Editing

Abstract: CRISPR-Cas9 has been touted for therapeutic genome editing, but limitations include inefficient correction of disease-causing mutations and off-target mutagenesis. In the latest issue of Cell, Liao et al. (2017) show that a modified version of CRISPR-Cas9 can target and activate key therapeutic genes in vivo without altering DNA sequence identity.

“…After the modification in Cas9 protein and the emergence of CRISPR/dCas9 technology, scientists start regulatory and reporter genes to investigate the ability of dCas9 for epigenome editing ( Zezulin and Musunuru, 2018 ). It was reported that the plant genome was modified by methylation and demethylation at target DNA and resultantly developed late-flowering phenotypes ( Adli, 2018 ).…”

“…Firstly, the endurance of gene activation or a repression effect mediated by CRISPR/Cas9 remains to be undetermined. It has been thought that epigenome-editing-induced gene activation or repression is short-term [ 90 , 91 ]. On the contrary, there was also evidence showing that a gene silencing effect mediated by the hit-and-run epigenome editing strategy could also be long-term and inheritable [ 31 ].…”

“…Third, environmentally induced epigenetic modification is transitory and can be reversed by further environmental changes, and is the basis for related recommendations about altering lifestyle and exposure to environmental factors in relation to methylation-based alterations in gene expression related to the development of cancer. Fourth, contemporary approaches to e-GE may only result in transient changes in target cells ( Zezulin and Musunuru 2018 ).…”
