Tumour Necrosis Factor Induced an Early Release of Superoxide and a Late Mitochondrial Membrane Depolarization in L929 Cells

Abstract: Tumour necrosis factor α (TNF) cytotoxicity is mediated, at least in part, by oxidative stress. One of the post-receptor events shortly after the addition of TNF is the generation of the superoxide anion (O2·). In the present study, we attempted to examine the role of O2· in the regulation of mitochondrial membrane potential (ΔΨm) and the release of cytochrome c (cyto c) in L929 cells after stimulation with TNF. Challenge of cells with TNF (50 ng/ml) resulted in an… Show more

“…Two sites of the mitochondrial respiratory chain have been identified as sources of ROS: complex 1 (NADH ubiquinone oxidoreductase) and complex 3 (the ubiquinone reductase site) (Liu et al, 2002). In the mitochondrial membrane, the ubiquinone reductase activity of complex 1 is sensitive to rotenone (Ko et al, 2001;Gyulkhandanyan et al, 2003). DPI inhibits the mitochondrial complex 1, especially between NADH and ferritin-sulfur clusters (Majander et al, 1994).…”

“…The antioxidant properties of NAC found in the present study may contribute directly to its inhibitory effects on NF-kB activation. Alternatively, NF-kB activation may result from the release of TNFa, which induces generation of reactive oxygen species [24] and has been found to be elevated in BALF from sensitised rats as early as 1 h after antigen challenge [25].…”

“…The mitochondrial membrane potential (¢"m) of cells treated with cytotoxic agents or medium alone was measured as previously described [19]. Briefly, cells after treatment were loaded with JC-1 (final concentration 10 ÌM ) at room temperature for 20 min.…”
