Tumor microenvironment and epithelial-mesenchymal transition in bladder cancer: Cytokines in the game?

Abstract: Bladder cancer (BlCa) is a highly immunogenic cancer. Bacillus Calmette-Guérin (BCG) is the standard treatment for non-muscle invasive bladder cancer (NMIBC) patients and, recently, second-line immunotherapies have arisen to treat metastatic BlCa patients. Understanding the interactions between tumor cells, immune cells and soluble factors in bladder tumor microenvironment (TME) is crucial. Cytokines and chemokines released in the TME have a dual role, since they can exhibit both a pro-inflammatory and anti-in… Show more

“…Additionally, intrinsic signaling pathways, which may be influenced by epigenetic changes and genetic mutations, could also lead to increased PD-L1 expression [22]. Furthermore, cytokines might influence the tumor microenvironment, regardless of BCG exposure [6,23]. However, our study clearly indicates that CPS levels show significantly greater changes in patients treated with BCG.…”

“…TME comprises various components, including tumor cells, immune cells, inflammatory cells, stem cells, vascular system, fibrous tissue, and extracellular matrix (ECM), and TME has been demonstrated to play a crucial role in the occurrence and progression of BLCA [ 20 ]. Given that CXCL10 may influence BLCA through multiple immune processes, we conducted an analysis of the role of CXCL10 in the TME.…”

“…Interestingly, we found that virtually all fibroblasts were positive for α-SMA, being classified as myofibroblast-like CAFs (myCAFs) [ 41 ]. MyCAFs play a crucial role in the tumor microenvironment by contributing to the deposition and organization of extracellular matrix components, including collagen and fibronectin, which provide structural support to the growing tumor mass in mouse subcutaneous tumor models [ 42 ]. In this line, the increase in the percentage of myCAFs in the treatment groups could be associated with a remodeling in the tumor’s extracellular matrix by treatments’ actions.…”
