TSU68 Prevents Liver Metastasis of Colon Cancer Xenografts by Modulating the Premetastatic Niche

Abstract: The aim of this study was to investigate the inhibitory effect of TSU68 [(Z)-5-[(1,2-dihydro-2-oxo-3H-indol-3-ylidene)methyl]-2,4-dimethyl-1H-pyrrole-3-propanoic acid; SU6668], an inhibitor of vascular endothelial growth factor receptor 2, plateletderived growth factor receptor B, and fibroblast growth factor receptor 1 (FGFR1), on colon cancer liver metastasis, and to test the hypothesis that TSU68 modulates the microenvironment in the liver before the formation of metastasis. First, we implanted the highly m… Show more

“…28, Figure 8C, and Supplemental Figure 7A). Not surprisingly, there was a significant overlap in up-and downregulated genes between pregnant mouse lungs and liver ( Figure 8D, Supplemental Figure 7B, and Supplemental Table 2), and several of the shared upregulated genes could be associated with infiltration of myeloid cells -possibly MDSCs.…”

“…In a nude mouse model of breast cancer, an increased infiltration of neutrophils increased invasiveness of the tumour, likely due to protease production (Yao et al 2007). In addition, the depletion of neutrophils in mouse models of fibrosarcoma and colon cancer significantly decreased metastasis in these animals (Tazawa et al 2003, Yamamoto et al 2008. The increase in neutrophil infiltration in endometrial adenocarcinoma induced by elevated chemokine expression has been described (Wallace et al 2009); however, the exact role that they are playing in endometrial adenocarcinoma is as yet unclear.…”

“…Some promising achievements regarding translation of exosomerelated metastasis targets have been documented. TSU68, a small molecule inhibiting VEGFR1, PDGFRβ and FGFR1 has been shown to suppress CXCL1/CXCR2-dependent niche formation in the liver by inhibition of the inflammatory response through reduction of CXCL1 and IL12 (137). Furthermore, antibodies directed against αv on breast cancer cells prevented metastasis to other organs, but not to the lung (138,139,140).…”
