Trypsin-Like Arginine Amidases Including Plasminogen and Plasmin in Human Seminal Plasma by Affinity Adsorption and Elution

Abstract: An enzyme preparation with affinity to a lysine column was detected from a DEAE-cellulose-adsorbed preparation of human seminal plasma containing plasminogen and plasmin. Two kinds of trypsin-like acidic arginine amidase activity with different affinity to lima bean trypsin inhibitor (LBTI) and aprotinin affinity column were detected from the DEAE-cellulose-adsorbed preparation after treatment of the lysine column. Two kinds of trypsin-like basic arginine amidase activity were also separated by the above-menti… Show more

“…We have separated some acidic and basic arginine amidases from human semen which contain a large amount oftrypsinlike proteinase inhibitor showing strong inhibition against the basic arginine amidases, in human semen [7,10,11], by using the LBTI and other affinity adsorption method. Some of their properties, including boar, were characterized.…”

“…amidase [10], tissue plasminogen activator (t-PA, EC 3.4.21.68) [18,19], prostate speciÂźc antigen (PSA) [14,20], and leucine aminopeptidase (EC 3.4.11.1)-like aryl amidases [15], have been found and separated from human semen (seminal plasma).…”

“…The proteinase inhibitors [3,6,81 and various proteinases have been detected in seminal plasma, including plasminogen/plasmin (EC 3.4.2 1.7) [9], tissue plasminogen activator (t-PA, EC 3.4.21.68) [2,16,181, tissue kallikrein (EC 3.4.21.35) [7,131, and other chymotrypsin (EC 3.4.21.1)-like [17] and trypsin (EC 3.4.21.4)-like basic arginine amidases [4, lo]. The physiological roles and significance of these proteinases and the proteinase inhibitors are currently under intense examination.…”
