To define and therapeutically target mechanisms that mediate nasopharyngeal carcinoma (NPC) metastasis, we have developed a unique orthotopic xenograft mouse model that accurately recapitulates the invasive and metastatic behavior of human disease. Based on clinical and laboratory evidence that the PI3K/Akt/mTOR axis is involved in aggressive NPC tumor behavior, we chose it as a therapeutic target to test the utility of our orthotopic system for evaluating the effectiveness of a targeted treatment for metastatic NPC. Demonstrated herein, we have shown that both the development and growth of metastatic lesions are markedly reduced by the mTOR inhibitor sirolimus. Thus, this orthotopic model provides a platform to study potential therapeutics for advanced NPC and demonstrates that targeting the PI3K/Akt/mTOR pathway is a promising intervention against disseminated disease.

<p class="abstract"><span lang="EN-IN">Nasopharyngeal carcinoma is one of the most difficult tumors to diagnose at early state and intracranial spread is common. We report the case of 45 years old Indian female with nasopharyngeal carcinoma, who presented with nasal obstruction and repeated cough and cold from last two months. X-ray PNS imaging revealed mild septal deviation to right side. Nasal endoscopy revealed lobulated greyish coloured mass involving both choana and mass protruded out on expiration. CT scan revealed soft tissue density in nasopharynx involving bilateral choanae. Excision mass done with presumptive diagnosis of lymphoma. Histopathological examination diagnosed non keratinizing undifferentiated nasopharyngeal carcinoma. Serum EBV-DNA levels were raised. Radiotherapy started immediately after diagnosis of NPC. Non-specific symptoms in this case, when followed up carefully diagnosed as NPC. Diagnosis is important for proper management.</span></p>

Trigeminal nerve palsy in nasopharyngeal carcinoma: Correlation between clinical findings and magnetic resonance imaging

Abstract: The incidence of MRI-evidenced trigeminal nerve involvement is high and often asymptomatic in NPC. Almost all patients with clinical trigeminal nerve palsies had MRI evidence of trigeminal nerve involvement in intracranial or orbital segment, especially in cavernous sinus.
