Treg: A Promising Immunotherapeutic Target in Oral Diseases

Abstract: With the pandemic of COVID-19, maintenance of oral health has increasingly become the main challenge of global health. Various common oral diseases, such as periodontitis and oral cancer, are closely associated with immune disorders in the oral mucosa. Regulatory T cells (Treg) are essential for maintaining self-tolerance and immunosuppression. During the process of periodontitis and apical periodontitis, two typical chronic immune-inflammatory diseases, Treg contributes to maintain host immune homeostasis and… Show more

“…Treg cells can control immune response in both effector and inductive sites, as represented in Figure 1. Foxp3+ Treg cells are activated in lymphoid structures under inflammatory conditions and then recruited to effector site by specific combinations of chemotactic molecules [23,24]. The CCL22-CCR4 axis seems to be particularly relevant to the recruitment of Tregs cells, as shown in mice periodontitis models [25].…”

“…Recent studies have focused on Th17 and Treg cells, which have opposite roles in the immune response in periapical lesions (Toledo et al, 2019). Treg and Th17 cells are thought to be important junctions between the immune system and bone (Zhang et al, 2021). According to a recent review, IL-17 and TNF-a secreted by Th17 cells promote RANKL expression and osteoclast differentiation, while TGF-b and IL-10 secreted by Treg cells inhibit this process (Wei et al, 2021).…”

“…Th17 cells are an important subset of effector T cells that are protective during extracellular bacterial and fungal invasion [90][91][92][93][94]. Treg cells are essential for maintaining self-tolerance and immunosuppression [95]. As discussed above, probably due to the reduction in oral clearance and local antibodies or antimicrobial peptides, several opportunistic pathogens are proportionally enriched in old mice.…”
