Trazodone: A Review of Its Pharmacological Properties and Its Off-Label Use in Dogs and Cats

Abstract: Trazodone is approved by the Food and Drug Administration (FDA) for the treatment of depression and anxiety in humans, it can perform these functions due to its selective serotonin receptor antagonism and reuptake inhibition properties. Trazodone has not been approved for animals by the FDA but it is legally prescribed by veterinarians as an offlabel drug for dogs and cats. This review describes trazodone's mechanism of action, pharmacokinetic properties and adverse effects and summarizes the benefits of trazo… Show more

“…In humans, trazodone has been shown to decrease plasma cortisol concentrations compared with placebo and is commonly prescribed for the treatment of anxiety, depression, and to facilitate sleep (7,32). While trazodone is not currently labeled for use in dogs, off-label use of trazodone is common for the treatment of anxiety disorders as well as to reduce the agitation and distress associated with post-surgery confinement and reduced exercise (33,34). In this experiment, treatment with trazodone lowered blood cortisol concentrations compared to all other treatments.…”

“…It is thought that the wide range of time to maximum concentration may be due to changes in gastric residence time in fed versus fasted dogs 2 . A previous study reported a maximum concentration within the range of 2.5–7.5 hours in fasted dogs 22,23 . Unfortunately, there are no studies that have investigated the metabolites of trazodone in the dog.…”

“…In veterinary medicine, trazodone (TRZ), another phenylpiperazine chemical derivative, has gained popularity as adjunctive off label medication for long term treatment of anxiety disorders in dogs (Gruen and Sherman, 2008) and as an anxiolytic for short-term management of patients after surgery (Jay et al, 2013;Chea and Giorgi, 2017). VLZ might have the potential to have better pharmacokinetic, pharmacodynamic and safety profiles than trazodone in the dog as theorized in human beings.…”
